单细胞测序技术在肿瘤浸润B细胞研究中的应用

摘要 肿瘤浸润B细胞(tumor-infiltrating B cells,TIL-Bs)已经被证明在调节肿瘤免疫方面起着关键作用,既可以抑制也可以促进抗肿瘤免疫的效应,需要对其进行深入分析以研究具体的效应功能。单细胞测序技术是对单个细胞的基因组、转录组及蛋白组进行高通量测序,利用其高分辨率对单个细胞的详细分子状态进行定义。因此,单细胞测序技术已经成为生命科学研究领域不可或缺的工具,可以全面分析不同类型肿瘤中B细胞的状态。本文从单细胞的角度阐述了TIL-Bs的异质性、优势细胞群体的状态并希望将对表型的表征转化为对预后以及治疗的研究,旨在认识TIL-Bs的生物学作用、肿瘤特异性抗体的功能,为靶向肿瘤的治疗提供一个新的角度。 Tumor-infiltrating B cells(TIL-Bs)have been shown to play a key role in modulating tumor immunity,both inhibiting and promoting the effects of anti-tumor immunity,and it needs to be analyzed in depth to investigate specific effector functions.Single-cell sequencing technology is a high-throughput sequencing of the genome,transcriptome and proteome of a single cell,using its high resolution to define the detailed molecular state of a single cell.Therefore,single-cell sequencing technology has become an indispensable tool in the field of life science research,which can comprehensively analyze the state of B cells in different types of tumors.This article expounds the heterogeneity of TIL-Bs from the perspective of single cells,the status of the dominant cell population,and hopes to transform the characterization of phenotype into prognosis and treatment research,aiming to understand the biological role of TIL-Bs and the function of tumor-specific antibodies,so as to provide a new perspective for targeted tumor therapy.
