PRRSV感染仔猪腹股沟淋巴结组织病理学观察及转录组分析

Histopathological observation and transcriptome analysis of inguinal lymph nodes inPRRSV-infected piglets

摘要 为探究猪繁殖与呼吸综合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)感染仔猪导致的腹股沟淋巴结的组织病理学变化及致病机制,对PRRSV感染仔猪腹股沟淋巴结进行组织病理学观察及转录组分析。结果显示,PRRSV感染仔猪腹股沟淋巴结肿胀出血,镜检观察可见淋巴组织结构模糊,淋巴细胞减少,出血;淋巴小结数量减少,界限不清,生发中心大量淋巴细胞坏死。基于Illumina测序平台,运用RNA-seq技术进行建库测序和生物信息分析,发现TLR8、CCL5、CCL8、IFNG、IRF5、TNFSF13B等炎性因子差异性表达,Toll样受体通路、NF-κB信号通路、TNF信号通路等通路被激活,提示这些细胞因子及信号通路参与PRRSV感染引起的淋巴结炎的病理变化过程。 To explore the histopathological changes and pathogenic mechanism of inguinal lymph nodes in piglets infected with porcine reproductive and respiratory syndrome virus(PRRSV),histopathological observation and transcriptome analysis of inguinal lymph nodes in PRRSV-infected piglets were carried out.The results showed that the inguinal lymph nodes were swollen and bleeding in PRRSV-infected piglets.Microscopic examination showed that the structure of lymphoid tissue was blurred,lymphocytes decreased and bleeding.The number of lymphoid nodules decreased with unclear boundary.A large number of lymphocytes in germinal center were necrotic.RNA-seq technology was used for library construction sequencing and bioinformatic analysis based on the Illumina sequencing platform.Several inflammatory factors including TLR8、CCL5、CCL8、IFNG、IRF5 and TNFSF13B were differentially expressed.Inflammatory signaling pathways such as Toll-like receptor pathway,NF-κB signaling pathway,TNF signaling pathway were activated.It shows that these cytokines and signaling pathways are involved in the pathological changes of lymphadenitis caused by PRRSV infection.
