正交试验优化齿瓣石斛多糖提取工艺

Optimization of Extraction Process of Polysaccharide from Dendrobium devonianum by Orthogonal Experiment

摘要 以水提法为基础,分别采用超声提取、水浴回流提取2种提取方式,研究料液比、提取温度、提取时间、提取次数4个变量对齿瓣石斛多糖提取率的影响,并利用正交试验优化齿瓣石斛多糖的提取工艺。用硫酸-蒽酮法检测多糖的含量,以多糖的提取率作为评价指标,判断最佳提取工艺。结果表明,齿瓣石斛多糖的最佳提取方式为超声提取,提取条件为料液比1︰40 (g/mL)、提取温度100℃、提取时间4 h、提取次数2次,此时提取率为30.66%。提取方式、料液比、提取温度、提取时间、提取次数均会对样品提取多糖的含量有显著影响,影响程度依次为提取时间>提取次数>料液比>提取温度>提取方式。 Based on the water extraction method,orthogonal experiments were used to optimize the extraction process of polysaccharides from Dendrobium dentatum.The effects of material liquid ratio,extraction temperature,extraction time and extraction times on the extraction rate of polysaccharides were studied using ultrasonic extraction and water bath reflux extraction.The content of polysaccharide was measured by anthrone-sulfuric acid method.The extraction yield of polysaccharide was used as the evaluation index to optimize the extraction process.The results showed that the optimum extraction method of Dendrobium dentatum polysaccharide was ultrasonic extraction,and the extraction condition was solid-liquid ratio of 1:40(g/mL),extraction temperature of 100℃,extraction time of 4 h and the extraction times of 2 times.Under this condition,the extraction rate of crude polysaccharide from Dendrobium devonianum was 30.66%.Extraction method,solid-liquid ratio,extraction temperature,extraction time and extraction times had a significant impact on the content of extracted polysaccharide.The order of influence was that extraction time>extraction times>solid-liquid ratio>extraction temperature>extraction method.
