摘要
为探讨新疆枸杞作为制备伤口敷料负载物的潜能,利用单因素法提取枸杞中的活性物质,利用DPPH法、抑菌试验检测枸杞提取物的抗氧化活性、抑菌性能,并利用荧光染色试验及扫描电子显微镜(SEM)对枸杞提取物的抑菌机制进行初步分析。此外,利用CCK-8法检测枸杞提取物的细胞毒性。结果表明,枸杞提取物的最佳提取条件为:在70℃水浴中用80%乙醇以料液比1︰8 (g/mL)静置90 min,此时提取物中的黄酮含量最高,为15.52 mg/g。抗氧化活性检测结果表明,枸杞提取物能有效清除DPPH自由基,提取物浓度2.5 mg/mL时,清除效果接近同等浓度下的VC溶液。抑菌试验表明,枸杞提取物浓度分别为0.2和0.1 g/mL时,其对E. coli和S. aureus有良好的抑制作用,通过对细菌的荧光染色发现,枸杞提取物能够通过破坏细菌细胞膜抑菌。CCK-8试验结果表明,枸杞提取物对L929细胞没有毒性并能够促进其增殖。枸杞提取物具有良好的抑菌、抗氧化活性,且对细胞无毒,表现出良好的生物活性,在治疗皮肤伤口愈合等医用材料方面具有很大潜力。
In order to explore the potential of Xinjiang Lycium barbarum as a load for the preparation of wound dressings,the extraction of Lycium barbarum was carried out using the single-factor method,the antioxidant activity and antibacterial properties of Lycium barbarum extracts were examined using the DPPH method and the inhibition assay,and the preliminary analysis of the inhibition mechanism of Lycium barbarum extracts was carried out using fuorescence staining experiments as well as scanning electron microscopy(SEM).In addition,the cytotoxicity of Lycium barbarum extract was detected using CCK-8 method.The results showed that the optimal extraction condition for the extract of Lycium barbarum was as follows:the highest flavonoid content of 15.52 mg/g was obtained after 90 min of resting in a water bath at 70℃ with 80%ethanol at a stock-to-liquid ratio of 1:8(g/mL).The results of the antioxidant activity assay showed that the extract of Lycium barbarum was able to effectively scavenge DPPH radicals,and the scavenging effect was similar to that of VC at the same concentration of 2.5 mg/mL.The effect was close to that of VC solution at the same concentration.Bacterial inhibition experiments showed that when the concentration of Lycium barbarum extract was 0.2 and 0.1 g/mL,respectively,it had good inhibitory effect on E.coli and S.aureus,and by fluorescence staining of bacteria,it was found that Lycium barbarum extract could inhibit bacteria by disrupting bacterial cell membrane.The results of CCK-8 experiment showed that LBP extract was not toxic to L929 cells and could promote their proliferation.The extract has good antibacterial and antioxidant activities,it is non-toxic to cells,showing good bioactivity,and it has great potential in the treatment of skin wound healing and other medical materials.
