基于植物乳杆菌GL-5的炎性肠病全营养配方食品对溃疡性结肠炎小鼠疾病的改善

Improvement of ulcerative colitis in mice induced by Lactobacillus plantarum GL-5 based total nutrition formula for inflammatory bowel disease

摘要 目的探讨植物乳杆菌GL-5炎性肠病全营养配方食品对葡聚糖硫酸钠诱导的溃疡性结肠炎小鼠疾病的改善作用。方法将78只C57BL/6雌性小鼠随机分为正常组、模型组、配方组、安素组、美沙拉嗪组和联合治疗组,适应7 d后,除正常组给予纯化水外,其余各组给予3.5%葡聚糖硫酸钠水溶液造模7 d,造模成功并干预10 d后处死。试验期间每天记录小鼠体重、摄食量、饮水量和不良事件发生情况,干预结束取血清测生理生化指标评估安全性,其间进行2次疾病活动指数评分和组织病理学评分评估临床效果。结果干预后,配方组、安素组、联合治疗组的体重恢复速度和最终体重优于美沙拉嗪组和模型组;配方组和联合治疗组的摄食和饮水量显著恢复,且两组的不良事件发生率低于模型组;各组干预后疾病活动指数评分均较造模期降低,与模型组相比,配方组、安素组、联合治疗组评分差异均有统计学意义(P<0.05);配方组和联合治疗组的近端结肠切片和病理学评估表现更为良好(P<0.05)。结论在溃疡性结肠炎治疗过程中,尽早地给予含辅助治疗的营养支持或可能改善疾病的转归。 Objective To study the improvement effect of Lactobacillus plantarum GL-5 whole nutrient formula on ulcerative colitis mice induced by dextran sulfate sodium salt.Methods 78 C57BL/6 female mice were randomly divided into 6 groups:normal group,model group,self-made formula group,ansu group,mesalazine group and combined treatment group.After 7 days of adaptation,except for group N,the mice were given purified water,and the other groups were given 3.5%dextran sulfate sodium salt aqueous solution for 7 days.The body weight,food intake,water intake,and adverse events of the mice were collected daily during the whole experiment.At the end of the intervention,serum was collected to measure physiological and biochemical indicators to evaluate the safety of the mice.At the end of the modeling and the end of the intervention,disease activity index was scored twice to evaluate the clinical effect.Results After intervention,the weight recovery rate and final weight of homemade recipe group,ansu group and combination group were better than those of mesalazine group and model group;The intake of food and water in the homemade recipe group and the combination group were significantly restored,and the incidence of adverse events in the two groups was lower than that in the model group;After intervention,the score of disease activity index after intervention in each group was lower than that in the modeling period.Compared with the model group,the scores of the homemade recipe group,the ansu group and the combination group were significantly different(P<0.05);The proximal colon section and pathological evaluation of the homemade recipe group and the combination group were better(P<0.05).Conclusion Early administration of nutritional support with adjuvant therapy in the treatment of ulcerative colitis may improve the outcome of the disease.
