摘要
目的本研究目的在于通过临床病理学研究方法,探讨METTL3在喉癌中的表达情况及与喉癌发生发展的关系。方法首先通过细胞实验验证METTL3在喉癌中的作用,CCK-8检测METTL3敲除实验后,喉癌细胞的增殖活性。另外收集被诊断为喉癌的患者79例,详细记录患者基本信息。利用qPCR检测组织中METTL3的mRNA的表达,并且进行Western Blot实验检测相应蛋白表达水平。采用皮尔逊卡方检验,分析临床参数与METTL3基因表达水平之间的相关性。采用斯皮尔曼检验,对临床资料和METTL3进行相关性分析。采用单因素Cox回归分析和多因素Cox回归分析计算患者特征与METTL3的风险值。对患者进行随访,并进行Kaplan-Meier生存分析。结果细胞实验结果显示,将喉癌细胞中METTL3沉默后,细胞增殖、侵袭和转移能力下降(P<0.05)。在喉癌组织水平中,RT-qPCR实验和Western Blot实验结果表明,METTL3在喉癌组织中显著上调。统计分析得出,METTL3表达水平与生存时间(P<0.001)显著相关。单因素Cox回归得出METTL3表达水平(HR=28.146,95%CI:11.644-68.035,P<0.001)与患者的生存时间有明显相关性。多变量Cox比例回归分析的结果显示,METTL3表达水平(HR=44.385,95%CI:15.934-123.637,P<0.001)与患者的生存时间仍然有明显相关性。通过Kaplan Meier方法分析得出,与低表达METTL3的喉癌患者相比,高表达METTL3的喉癌患者生存较差(HR=44.385,P<0.001)。结论METTL3在喉癌中可能发挥原癌基因的功能,与喉癌患者的生存具有密切相关性,进而为临床喉癌的治疗提供新的思路及策略,这将为喉癌的防治开辟一个可能的新靶点。
Objective The aim of this study is to investigate the expression of METTL3 in laryngeal cancer and its relationship with the occurrence and development of laryngeal cancer by using clinicopathological methods.Methods The role of METTL3 in laryngeal cancer was verified by cell experiments,and CCK-8 was used to detect the proliferation activity of laryngeal cancer cells after METTL3 knockdown experiment.In this study,79 patients diagnosed with laryngeal cancer were collected and their basic information was recorded in detail.qPCR was used to detect the mRNA expression of METTL3 in tissues,and Western Blot was used to detect the corresponding protein expression level.Pearson chi-square test was used to analyze the correlation between clinical parameters and METTL3 gene expression level.Spearman test was used to analyze the correlation between clinical data and METTL3.Univariate and multivariate Cox regression analyses were used to calculate risk values of METTL3on patient characteristics.Patients were followed up and Kaplan-Meier survival analysis was performed.Results The results of cell experiments showed that after silencing METTL3 in laryngeal cancer cells,cell proliferation,invasion and metastasis were decreased(P<0.05).At the tissue level of laryngeal cancer,the results of RT-qPCR and Western Blot showed that METTL3 was significantly up-regulated in laryngeal cancer tissues.Statistical analysis showed that METTL3 expression was significantly correlated with survival time(P<0.001).Univariate Cox regression showed that METTL3 expression level(HR=28.146,95%CI:11.644-68.035,P<0.001)was significantly correlated with patient survival time.MultivariateCox proportional hazard analysis showed that METTL3 expression level H(R=44.385,95%CI:15.934-123.637,P<0.001)was still significantly correlated with patient survival time.Kaplan-meier analysis showed that patients with high expression of METTL3 had poor survival compared with patients with low expression of METTL3(HR=44.385,P<0.001).Conclusion METTL3 may play the role of proto-oncogene in laryngeal cancer,which is closely related to the survival of laryngeal cancer,thus providing new ideas and strategies for the treatment of laryngeal cancer,which will open up a possible new target for the prevention and treatment of laryngeal cancer.
