甲状腺癌关键致病基因筛选与预后分析及治疗中药的预测研究

Screening and Prognostic Analysis of Key Pathogenic Genes of Thyroid Carcinoma and Prediction of Traditional Chinese Medicine for Treatment

摘要 运用生物信息学方法,分析甲状腺癌的核心致病基因和关键预后基因,并预测可用于甲状腺癌靶向治疗的中药及活性成分。从GEO数据库下载甲状腺癌和健康人的基因芯片数据,用GEO2R工具,分析甲状腺癌和正常甲状腺组织间的显著性差异表达基因(DEGs);利用Metascape和GSEA数据库,分别对显著性DEGs进行GO和信号通路分析;采用GEPIA数据库,对显著性DEGs进行生存分析,筛选并验证甲状腺癌关键致病基因;运用TIMER数据库,探究关键致病基因表达与免疫浸润的关系;将关键基因导入Coremine数据库,挖掘可用于甲状腺癌靶向治疗的中药;通过CTD数据库,预测可用于甲状腺癌靶向治疗的中药活性成分。共筛选出393个显著性DEGs。显著性DEGs主要集中于肌肉收缩、对激素的反应、炎症反应等生物学过程,参与组成收缩纤维、肌质网、细胞间连接等细胞成分,影响免疫受体活性、信号素受体活性、趋化因子受体结合等分子功能。信号通路集中于细胞粘附分子、趋化因子信号通路、细胞因子受体相互作用、FcγR介导的吞噬作用等信号通路。对甲状腺癌预后有显著影响的DEGs为CLDN1、CXCL10、APOC1、CDH3、MYOT和AKR1C3,这些基因的表达与免疫浸润均显著相关。预测可用于甲状腺癌治疗的中药有黄芪、黄芩、半枝莲、野菊花、杜仲等。中药活性成分主要包括阿魏酸、小檗碱、芹菜素、木犀草素、山萘酚、黄芩素、人参皂苷等。本研究探寻到影响甲状腺癌发病及预后的关键基因,并进一步预测了可用于甲状腺癌治疗的中药,结果可为甲状腺癌的诊治提供新的分子生物标志物,并为甲状腺癌的临床治疗提供新思路。 The study analyzes the core pathogenic genes and key prognostic genes of thyroid cancer by bioinformatics and predicts Chinese medicine and its active components for targeted therapy of thyroid cancer.Screening and downloading gene chip data for thyroid cancer and healthy people from GEO database,the study analyzes the differential expression genes(DEGs)between thyroid cancer and normal thyroid tissues by GEO2R;using Metascape database and GSEA database,the study does GO enrichment analysis and signal pathway analysis of DEGs;using GEPIA database,the study analyzes the survival of DEGs,and screens and verifies the key pathogenic genes of thyroid cancer;using TIMER database,the study explores the relationship between the expression of key pathogenic genes and immune infiltration;importing key genes into Coremine database,the study mines traditional Chinese medicine for targeted therapy of thyroid cancer;the study predicts the active components of traditional Chinese medicine that can be used for targeted treatment of thyroid cancer through CTD database.A total of 393 DEGs are selected.Indigenous DEGs are mainly concentrated in biological processes,such as muscle contraction,response to hormones,and inflammatory response,etc.They are involved in the composition of contraction fibers,sarcoplasmic reticulum,intercellular junctions and other cellular components,affecting the activity of immune receptors,the activity of signaling receptors,and the binding of chemokine receptors.Signaling pathways focus on cell adhesion molecules,chemokine signaling pathways,cytokine receptor interaction,FcγR-mediated phagocytosis and other signaling pathways.The dominant DEGs affecting the prognosis of patients with thyroid cancer are CLDN1,CXCL10,APOC1,CDH3,MYOT and AKR1C3.The expression of these genes is significantly correlated with immune infiltration.The predicted Chinese medicines for thyroid cancer treatment are Astragalus membranaceus,Scutellaria baicalensis,Scutellaria barbata,Chrysanthemum indicum,and Eucommia ulmoides,etc.The active ingredients of traditional Chinese medicine mainly include ferulic acid,berberine,apigenin,luteolin,kaempferol,baicalein,and ginsenoside,etc.This study explores the key genes affecting the pathogenesis and prognosis of thyroid cancer,and further predicts the Chinese medicine that can be used for the treatment of thyroid cancer,which can provide new molecular biomarkers for the diagnosis and treatment of thyroid cancer,and provide new ideas for the research and development of related Chinese medicine.
