摘要
NT5C2所编码的蛋白为细胞质5’-核苷酸酶Ⅱ(cN-Ⅱ),该酶具有催化核苷单磷酸酯去磷酸化作用,主要参与细胞内嘌呤核苷酸库的调节。高表达cN-ⅡmRNA与急性白血病(AL)患者预后较差有关,获得性NT5C2突变会阻碍DNA和巯鸟嘌呤核苷酸(TGN)结合物的形成,导致6-巯基嘌呤(6-MP)耐药。此外,NT5C2与2型糖尿病、精神分裂症等多种疾病的发生风险都具有关联性。
Cytosolic 5’-nucleotidaseⅡ(cN-Ⅱ),the protein encoded by NT5C2,can catalyze the dephosphory-lation of nucleoside monophosphate and is mainly involved in the regulation of purine nucleotide pool in cells.High expression of cN-ⅡmRNA is associated with poor prognosis of patients with acute leukemia(AL).Acquired NT5C2 mutation will hinder the formation of DNA and thioguanine nucleotide(TGN)conjugate,resulting in 6-mercaptpurine(6-MP)resistance.In addition,NT5C2 is a risk factor associated with typeⅡdiabetes,schizophrenia and other diseases.
