番茄髓部坏死病病原菌分离鉴定及药剂筛选

Identification of causal agent of tomato pith necrosis and screening of fungicides

摘要 该研究对2020年浙江设施栽培的番茄进行病害调查,利用柯赫氏法则进行病原菌的致病性测定,采用形态观察、革兰氏染色法、脂肪酸含量测定及16Sr RNA-gyr B-rpoB-rpo D四基因联合系统发育分析等方法对分离的病原菌进行种属鉴定,并进行药剂室内毒力测定。该研究表明番茄髓部坏死病有明显的发病中心,设施大棚中易传播扩散;经形态学、生理生化和分子鉴定等方法确认该病原菌为绿黄假单胞菌(Pseudomonas viridiflava);中生菌素对番茄髓部坏死病菌的抑制效果较好,其EC50仅0.106 mg/L,是较为理想的杀菌剂。 2020,the tomato disease field investigation in greenhouses of Zhejiang province is conducted.Koch's rule is used to determine the pathogenicity of pathogens.Morphological observation,Gram staining,fatty acid profiling and 16S rRNA-gyrB-rpoB-rpoD four genes combined phylogenetic analysis are used to identify the species of isolated pathogens.The toxicity of the reagent is measured in laboratory.There are obvious infection centers of tomato pith necrosis,and it is easy to spread in the greenhouse.The pathogen is clearly supported as Pseudomonas viridiflava by morphological,physiological,biochemical and molecular identification.As EC50 of the inhibitory effect of Zhongshengmycin is only 0.106 mg/L,it would be an ideal fungicide for tomato pith necrosis.
