黄连素通过TLR4途径对大鼠脊髓损伤后神经功能的影响及相关机制研究

摘要 目的探讨黄连素通过Toll样受体4(Toll-like receptor 4,TLR4)途径对大鼠脊髓损伤(spinal cord injury,SCI)后神经功能的影响及相关机制。方法将36只SD大鼠随机分为对照组、SCI组和SCI+黄连素组各12只。通过击打的方法建立SCI模型,对照组仅暴露脊椎。SCI+黄连素组大鼠通过黄连素灌胃(5 mg/100 g)。检测和比较各组BBB评分、血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)、脊髓组织中凋亡蛋白和TLR4通路水平。结果建模后7 d,各指标如下:SCI组的BBB评分<SCI+黄连素组<对照组,差异有统计学意义(P<0.05)。SCI组血清TNF-α和IL-6水平>SCI+黄连素组>对照组,差异有统计学意义(P<0.05)。SCI组的Bax、caspase3、TLR4、MyD88水平>SCI+黄连素组>对照组(P<0.05),而SCI组的Bcl2水平<SCI+黄连素组<对照组(P<0.05),差异均有统计学意义。结论黄连素可以通过抑制TLR4通路和炎性因子的水平抑制脊髓中神经元凋亡,从而保护SCI大鼠模型的神经功能。 Objective To investigate the effect of berberine on the neural function after spinal cord injury(SCI)in rats via Toll-like receptor 4(TLR4)pathway and related mechanisms.Methods A total of 36 SD rats were randomly divided into control group,SCI group and SCI+berberine group(n=12).The SCI model was established by beating,and the control group only exposed the spine.Rats in the SCI+berberine group were intragastrically given berberine(5 mg/100 g).BBB scores,serum tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),apoptotic protein and TLR4 pathway in spinal cord tissues were detected and compared.Results Seven days after modeling,the indicators were as follows:the BBB score of SCI group<SCI+berberine group<control group,with statistically significant difference(P<0.05),serum TNF-αand IL-6 levels of SCI group>SCI+berberine group>control group(P<0.05),there were statistically significant differences.The Bax,caspase3,TLR4,MyD88 levels in SCI group>SCI+berberine group>control group(P<0.05),while the level of Bcl2 in SCI group<SCI+berberine group<control group(P<0.05),with significant differences.Conclusion Berberine can inhibit neuronal apoptosis in the spinal cord by inhibiting the TLR4 pathway and the levels of inflammatory factors,thereby protecting the nerve function of the SCI rats.
