陈皮挥发油GC-MS分析与橙皮苷含量测定

摘要 目的:分析陈皮挥发油中化学成分并测定橙皮苷的含量。方法:以陈皮为主要研究实验对象,采用水蒸汽蒸馏法和溶剂热回流提取法分别提取陈皮挥发油及提取物,用气相色谱-质谱联用(GC-MS)方法和高效液相色谱(HPLC)法分别对其主要的化学成分以及橙皮苷的含量进行鉴定和分析。结果:通过分析得出陈皮挥发油中D-柠檬烯成分含量相对较高,占总量的94.15%,此外含量较高的还有γ-萜品烯和β-月桂烯,其相对含量为3.27%和1.20%。而橙皮苷在12.5~200μg/mL范围内的峰面积与进样浓度线性关系良好(R2=0.998),平均加样回收率为99.32%,其相对标准偏差(RSD)为2.47%。结论:采用高效液相色谱法测定陈皮中橙皮苷的含量,重现性和稳定性较好,操作简便,可作为快速、灵敏、准确检测橙皮苷的一种方法。 Objective:To analyze the chemical constituents of volatile oil and determine the content of hesperidin in Citrus reticulata pericarpium.Methods:Tangerine peel was the main research object.Volatile oil and extracts were extracted by steam distillation and solvent thermal reflux extraction,respectively.The chemical composition of volatile oil were determined by gas chromatography‑mass spectrometry(GC-MS)method,and the content of hesperidin were identified by high performance liquid chromatography(HPLC)method.Results:The content of D-limonene was relatively high in the volatile oil of tangerine peel,accounting for 94.15%of the total.In addition,the higher content ofγ-terpinene andβ-myrcene had the relative content of 3.27%and 1.20%.The peak area of hesperidin ranging from 12.5 to 200μg/ml was linearly related to the injection concentration(R2=0.998).The average sample recovery rate was 99.32%,and the relative standard deviation(RSD)was 2.47%.Conclusion:The content of hesperidin in tangerine peel was determined by HPLC,with good reproducibility,stability,and easy operation.It could be used as a method for rapid,sensitive and accurate detection of hesperidin.
