摘要
目的探讨核仁和纺锤体相关蛋白1(NUSAP1)在肺腺癌组织中的表达及临床意义。方法通过检索Oncomine、GEPIA、UALCAN、TIMER、STRING等数据库,分析肺腺癌NUSAP1基因与蛋白表达情况、NUSAP1表达对患者预后的影响、NUSAP1基因与肿瘤免疫细胞浸润的关系,以及蛋白网络图富集参与的生理过程等。结果NUSAP1在肺腺癌组织中高表达,基因表达越高患者预后越差,差异有统计学意义(P<0.05);NUSAP1在肺腺癌组织中表达水平与肿瘤微环境中B淋巴细胞、CD4^(+)T淋巴细胞、树突细胞均呈负相关(P<0.05),与CD8^(+)T淋巴细胞、巨噬细胞、中性粒细胞均呈正相关(P<0.05);NUSAP1主要参与调节细胞周期、有丝分裂细胞周期的正调控、核分裂等生物学过程。结论NUSAP1在肺腺癌组织中高表达,并与肺腺癌的发生、发展相关,可能作为肺腺癌诊断及预后评估的生物标志物。
Objective To investigate the expression and clinical significance of nucleolar and spindle associated protein 1(NUSAP1)in lung adenocarcinoma.Methods By searching Oncomine,GEPIA,UALCAN,TIMER,STRING and other databases,the expression of NUSAP1 gene and protein in lung adenocarcinoma,the influence of NUSAP1 expression on the prognosis of patients,the relationship between NUSAP1 gene and tumor immune cell infiltration,and the physiological process involved in protein network pattern enrichment were analyzed.Results NUSAP1 was highly expressed in lung adenocarcinoma tissues,and the higher the gene expression was,the worse the prognosis was,the difference was statistically significant(P<0.05).The expression level of NUSAP1 in lung adenocarcinoma was negatively correlated with B lymphocytes,CD4^(+)T lymphocytes and dendritic cells in tumor microenvironment(P<0.05),and positively correlated with CD8^(+)T lymphocytes,macrophages and neutrophils(P<0.05).NUSAP1 was mainly involved in the regulation of cell cycle,positive regulation of mitotic cell cycle,nuclear division and other biological processes.Conclusion NUSAP1 is highly expressed in lung adenocarcinoma tissues and is associated with the occurrence and development of lung adenocarcinoma,which may be used as a biomarker for diagnosis and prognosis evaluation of lung adenocarcinoma.
