lncRNA CASC9通过调节miR-125a-3p/VEGF对口腔鳞癌CAL-27细胞增殖、迁移和侵袭的影响

The impact of lncRNA CASC9 on the proliferation, migration and invasion of oral squamous cell carcinoma CAL- 27 cells by regulating miR- 125a- 3p/VEGF

目的:探究长链非编码RNA癌症易感性候选基因9(lncRNA CASC9)对口腔鳞癌(OSCC)CAL-27细胞增殖、迁移和侵袭的调控机制。方法:构建lncRNA CASC9敲低的CAL-27细胞,并在lncRNA CASC9敲低细胞系中进行microRNA-125a-3p(miR-125a-3p)的回复。检测细胞中lncRNA CASC9、miR-125a-3p和VEGF mRNA水平,检测细胞增殖、迁移及侵袭能力以及细胞中VEGF蛋白表达。结果:敲低lncRNA CASC9表达后,CAL-27细胞增殖活性、迁移与侵袭能力、VEGF mRNA和蛋白水平降低(P<0.05),miR-125a-3p水平升高(P<0.05)。敲低lncRNA CASC9可显著抑制细胞在体内移植瘤生长和血管生成(P<0.05)。下调miR-125a-3p表达可减弱lncRNA CASC9敲低对CAL-27细胞增殖、迁移和侵袭以及对体内移植瘤生长和血管生成的抑制作用(P<0.05)。结论:lncRNA CASC9可能通过调节miR-125a-3p/VEGF促进OSCC细胞增殖、迁移和侵袭。

Objective:To explore the regulatory mechanism of long non-coding RNA cancer susceptibility candidate 9(lncRNA CASC9)on the proliferation,migration and invasion of oral squamous cell carcinoma(OSCC)CAL-27 cells.Methods:lncRNA CASC9 knockdown CAL-27 cells were constructed by sh-CASC9,and the reversion of microRNA-125a-3p(miR-125a-3p)was performed in the lncRNA CASC9 knockdown cells by miR-125a-3P inhibitor.lncRNA CASC9,miR-125a-3p and VEGF mRNA levels in the cells were examined,cell proliferation,migration and invasion ability,the expression of VEGF proteins in the cells were tested by MTT assay,Transwell assay,in vivo experiment in nude mice,RT-qPCR and Wester blot,respectively.Results:After lncRNA CASC9 knockdown,the proliferation,migration and invasion abilities,VEGF mRNA and protein levels in CAL-27 cells were decreased(P<0.05),and the miR-125a-3p level was increased(P<0.05).Knockdown of lncRNA CASC9 inhibited the growth and angiogenesis of CAL-27 xenografts in vivo(P<0.05).Down-regulation of miR-125a-3p expression attenuated the inhibitory effects of lncRNA CASC9 knockdown on cell proliferation,migration and invasion,and in vivo xenograft growth and angiogenesis(P<0.05).Conclusion:LncRNA CASC9 may promote proliferation,migration and invasion of OSCC cells by regulating miR-125a-3p/VEGF.
