非小细胞肺癌组织中高尔基膜蛋白1表达水平及其与患者临床病理特征和免疫浸润的相关性 被引量:3

Expression level of GOLM1 in non-small cell lung cancer tissues and its correlation with clinicopathological characteristics and immune infiltration in patients

摘要 目的 探讨非小细胞肺癌(Non small cell lung cancer, NSCLC)组织中高尔基膜蛋白1(Golgi membrane protein 1,GOLM1)表达水平与患者临床病理特征及免疫浸润的相关性。方法 收集本院行手术治疗的NSCLC患者的癌旁组织与癌组织,qRT-PCR分析GOLM1在NSCLC中的表达水平。探讨GOLM1表达水平与NSCLC患者临床病理参数的关系。受试者工作特征曲线(Receiver operator characteristic curve,ROC)与Kaplan-Meier生存曲线分析GOLM1表达水平对诊断NSCLC的敏感性及患者生存率的影响。Logistic单因素与多因素分析影响患者生存率的危险因素。借助肿瘤免疫浸润评估资源(Tumor immune estimation resource, TIMER)数据库与肿瘤免疫系统相互作用(Tumor-immune system interactions, TISIDB)数据库分析GOLM1表达水平与NSCLC免疫细胞浸润水平的相关性。结果 与癌旁组织相比,GOLM1在癌组织中表达显著升高(P<0.05)。Fisher精确检验发现GOLM1表达水平与淋巴结分期、临床分期具有显著相关性(P均<0.05)。ROC曲线下面积为0.757 7,Kaplan-Meier分析显示GOLM1高表达的患者生存概率低于低表达的患者(P均<0.05)。Logistic回归检验显示肿瘤分期及GOLM1表达水平均是影响NSCLC患者生存的独立危险因素。GOLM1与肺腺癌中B细胞、树突状细胞的浸润水平呈负相关,GOLM1与肺鳞癌中CD8+T细胞、CD4+T细胞、巨噬细胞树突状细胞呈正相关(P均<0.05)。肺腺癌中GOLM1拷贝数变异对CD8+T细胞、CD4+T细胞、嗜中性粒细胞的浸润水平有显著影响,肺鳞癌中GOLM1拷贝数变异对CD8+T细胞、CD4+T细胞、巨噬细胞、嗜中性粒细胞、树突状细胞浸润水平有显著影响(P均<0.05)。结论 GOLM1表达水平与NSCLC患者临床病理特征、预后、免疫细胞浸润显著相关。 Objective The aim of the study was to investigate the relationship between the expression level of Golgi membrane protein 1(GOLM1)and the clinicopathological features and immune infiltration of non small cell lung cancer(NSCLC).Methods The paracancerous tissues and cancer tissues of the NSCLC patients who underwent surgical treatment in the hospital were collected,and the expression level of GOLM1 in NSCLC was analyzed by qRT-PCR.To investigate the relationship between the GOLM1 expression and the clinicopathological parameters of NSCLC patients.Receiver operator characteristic curve(ROC)and Kaplan-Meier survival curve was used to analyze the sensitivity of GOLM1 expression level in the diagnosis of NSCLC and the survival rate of the patients.The risk factors affecting the survival rate of the patients were analyzed by Logistic univariate and multivariate analysis.TIMER and TISIDB databases were used to analyze the correlation between GOLM1 expression level and immune cell infiltration in NSCLC.Results Compared with the adjacent tissues,the expression of GOLM1 was significantly increased in the cancer tissues(P<0.05).Fisher exact test showed that GOLM1 expression was significantly correlated with lymph node stage and clinical stage(all P<0.05).The area under the ROC curve was 0.7577.Kaplan-Meier analysis showed that the survival rate of the patients with high GOLM1 expression were lower than that of the patients with low GOLM1 expression(all P<0.05).Logistic regression analysis showed that tumor stage and GOLM1 expression was independent risk factor for survival of NSCLC patients.The expression of GOLM1 was negatively correlated with the infiltration level of B cells and dendritic cells in lung adenocarcinoma,and positively correlated with the infiltration level of CD8+T cells,CD4+T cells,macrophages and dendritic cells in lung squamous cell carcinoma(all P<0.05).Copy number variations of GOLM1 significantly affected the infiltration levels of CD8+T cells,CD4+T cells and neutrophils in lung adenocarcinoma.Copy number variations of GOLM1 significantly affected the infiltration levels of CD8+T cells,CD4+T cells,macrophages,neutrophils and DCS in lung squamous cell carcinoma(all P<0.05).Conclusion The expression level of GOLM1 is significantly correlated with clinicopathological characteristics,prognosis and immune cell infiltration of NSCLC patients.
