线粒体自噬基因E2F1与子宫内膜癌预后的相关性研究 被引量:1

摘要 目的:探讨线粒体自噬基因E2F1与子宫内膜癌(uterine corpus endometrial carcinoma,UCEC)预后的相关性。方法:R软件筛选肿瘤基因组图谱数据库UCEC以及正常组织中差异表达的线粒体自噬基因。单因素和多因素Cox回归分析发现E2F1与UCEC预后密切相关。Kaplan-Meier分析E2F1高危组和低危组生存时间的差异。GEPIA数据库探讨E2F1 mRNA在UCEC患者癌组织和正常组织表达情况。人类蛋白质图谱(the human protein at⁃las,HPA)在线数据库探讨E2F1蛋白在UCEC患者癌组织和正常组织表达情况。UALCAN数据库探讨E2F1表达与UCEC患者临床资料之间关系。结果:9个线粒体自噬基因在癌组织中低表达,5个线粒体自噬基因在癌组织中高表达。Kaplan-Meier分析E2F1表达低风险组患者的生存率优于高风险组(P<0.001)。GEPIA数据库发现E2F1 mRNA在UCEC癌组织中表达量高于癌旁正常组织(P<0.05)。HPA数据库发现E2F1蛋白在癌组织中高表达。E2F1表达量与UCEC患者组织学分型、肿瘤分期以及TP53突变密切相关(P<0.05)。结论:E2F1在UCEC中高表达,并且其表达水平与UCEC患者的不良预后密切相关。 Objective To investigate the relationship between mitophagy gene E2F1 and prognosis of uterine corpus endome⁃trial carcinoma(UCEC).Methods R software was used to screen tumor genome atlas database UCEC and differentially ex⁃pressed mitophagy genes in normal tissues.Univariate and multivariate Cox regression analysis revealed a close association between E2F1 and UCEC prognosis.Kaplan-Meier analysis was used to compare the survival time of UCEC patients with high and low E2F1 expression levels,and GEPIA database was used to investigate the expression of E2F1 mRNA in cancer tissues and normal tissues of UCEC patients.Furthermore,the human protein atlas(HPA)online was used to examine the expression of E2F1 protein in cancer tissues and normal tissues of UCEC patients.Lastly,we explored the correlation be⁃tween E2F1 expression and clinical data of UCEC patients using the UALCAN database.Results Nine mitochondrial auto⁃phagy genes were found to be downregulated in tumor tissues,while five genes were upregulated.Kaplan Meier analysis showed that the survival rate of patients in the low-risk group with E2F1 expression was better than that in the high-risk group(P<0.001).The GEPIA database found that the expression level of E2F1 mRNA in UCEC cancer tissue was higher than that in normal tissue adjacent to cancer(P<0.05).The HPA database found that E2F1 protein was highly expressed in cancer tissue.The expression level of E2F1 was closely related to the histological classification,tumor staging,and TP53 mutation in UCEC patients(P<0.05).Conclusion E2F1 is highly expressed in UCEC,and its expression level is closely related to the poor prognosis of patients with UCEC.
