摘要
以白及为原料,水提醇沉法提取白及多糖(Bletilla Striata polysaccharide,BSP),采用响应面法优化BSP脱蛋白后多糖保留率,并分析其光谱性质及电镜图。结果表明,BSP的最佳提取工艺为脱蛋白次数5次,Sevage试剂体积10 mL,振荡时间6 min,该条件下BSP得率为30.61%。各单因素对BSP得率的影响顺序为振荡时间>脱蛋白次数>Sevage试剂。红外光谱分析显示BSP具有典型的糖类特征吸收峰;同时电镜结果表明脱蛋白后的BSP结构致密,分子间的交联作用强。该结果为BSP的提取纯化提供实验支撑,可为BSP作为药物或辅料的后续研究提供参考。
The water extraction and alcohol precipitation method was used to optimize the extraction process of Bletilla Striata polysaccharide(BSP)by response surface methodology,and the spectral properties and electron micrographs were analyzed.The results showed that the optimal extraction process of BSP was as follows:the number of deproteinization was 5 times,the volume of Sevage reagent was 10 mL,and the shaking time was 6 min.Under these conditions,the yield of polysaccharide was 30.61%.The order of the influence of each single factor on the yield of BSP was shaking time>times of deproteinization>volume of Sevage reagent.Infrared spectral analysis showed that BSP had typical absorption peaks of carbohydrates.At the same time,the electron microscope results showed that the surface morphology of BSP was relatively uniform,and the spherical aggregate structure was compact.The results provide a basis for the extraction and purification of BSP,and support for subsequent research of BSP as a drug or excipient.
