尾穗苋体外细胞毒活性成分的研究

摘要 目的 探究尾穗苋中具有体外细胞毒活性的成分。方法 采用硅胶柱层析、Sephadex LH-20柱层析和高效液相色谱等方法对尾穗苋70%乙醇粗提物进行分离纯化,所得单体化合物的结构经多种核磁共振谱、熔点及高分辨质谱数据鉴定。采用MTT法对单体化合物进行体外细胞毒活性研究。结果 从尾穗苋中分离和鉴定了10个化合物,分别是:木犀草素-7-O-葡萄糖苷(1)、木犀草素(2)、1H-咪唑并[4,5-D]哒嗪-2-胺(3)、尿嘧啶核苷(4)、胸腺嘧啶核苷(5)、2β,3β-二羟基-30-降冰油烷-12,20(29)-二烯-23,28-二甲酸-28-O-β-D-吡喃葡萄糖酯(6)、齐墩果酸3-O-β-D-葡萄糖苷-28-O-β-D-吡喃葡萄糖酯苷(7)、2β-羟基齐墩果酸-3-O-β-D-葡萄糖苷-28-O-β-D-吡喃葡萄糖酯苷(8)、3-O-β-D-吡喃葡萄糖基-2β,3β-二羟油酸-12-烯-23,28-二甲酸-28-O-β-D-吡喃葡萄糖基酯苷(9)、3-氧代-α-紫罗兰醇-β-D-吡喃葡萄糖苷(10)。木犀草素(2)对人肝癌细胞系HepG2和SMMC-7721均有显著的体外细胞毒活性,其IC_(50)值分别为13.36±1.01μg/ml和16.70±1.15μg/ml。结论化合物1~5为首次从该植物中分离到,化合物2具有较强的体外细胞毒活性。 Abstrat:Objective To investigate the cytotoxic active constituents of Amaranthus caudatus L.in vitro.Methods The 70 % ethanol extract of A.caudatus was separated and purified by various column chromatographic methods such as silica gel,Sephadex gel LH-20,and HPLC.The structures of all compounds were elucidated by extensive NMR and/or melting point data in conjunction with HR-ESI-MS data.Then,the cytotoxic activity of the monomer compound in vitro was studied by MTT method.Results(1) Ten compounds were separated and identified as:luteolin-7-O-glucoside(1),luteolin(2),zarzissine(3),uridine(4),thymidine(5),2β,3β-dihydroxy-30-norolean-12,20(29)-diene-23,28-dioic acid-28-O-β-D-glucopyranosyl ester(6),3-O-β-D-glucopyranosyl-oleanolic acid-28-O-β-D-glucopyranosyl ester(7),3-O-β-D-glucopyranosyl-2β-hydroxyolean-olic acid-28-O-β-D-glucopyranossyl ester(8),3-O-β-D-glucopyranosyl-2β,3β-dihydroxyolean-12-ene-23,28-dioic acid-28-O-β-D-glucopyranosyl ester(9),and 3-oxo-4-hydroxy-β-ionyl-9-O-β-D-glucopyranoside(10),respectively.Luteolin(2) has significant cytotoxic activity against human hepatoma cell lines Hep G2 and SMMC-7721 in vitro,with the IC_(50) values of 13.36±1.01 μg/ml and 16.70±1.15 μg/ml,respectively.Conclusion Compounds 1-5 are obtained from the title plant for the first time.Compound 2 exhibits significant cytotoxic activity.
