基因编辑技术在谷氨酸棒杆菌中的应用研究进展

摘要 谷氨酸棒杆菌(Corynebacterium glutamicum)是一种绿色高效的底盘微生物,可用于生产大宗氨基酸、精细化学品、营养保健品等众多高附加值产品。基因编辑技术作为一种有效的调控菌株代谢网络的手段,在谷氨酸棒杆菌合成生物学研究、理性改造高产菌株等方面发挥着巨大潜力。然而,谷氨酸棒杆菌的遗传操作仍存在挑战。该文综述了谷氨酸棒杆菌常用基因编辑技术,详细阐述了传统基因编辑技术、规律成簇间隔短回文重复序列及其相关的蛋白(CRISPR/Cas)介导的基因编辑技术及CRISPR干扰(CRISPRi)技术的作用机制及其在谷氨酸棒杆菌中的应用研究进展。旨在为基因编辑技术在谷氨酸棒杆菌菌株设计和生产高附加值产品方面提供依据。 Corynebacterium glutamicum is a kind of green and efficient bacterium,which can be used to produce many high-value-added products,such as bulk amino acids,fine chemicals,nutrition and health products,and so on.As an effective means to regulate the metabolic network of strains,gene editing technology plays a great potential in the study of synthetic biology of C.glutamate and rational transformation of high-yield strains and so on.However,the genetic manipulation of C.glutamicum remains challenging.The common gene editing techniques of C.glutamicum were reviewed in this paper.The mechanism of traditional gene editing techniques,clustered regularly interspaced short palindromic repeats-associated protein(CRISPR/Cas)mediated gene editing techniques and CRISPR interference(CRISPRi)and their application in C.glutamate were reviewed in detail.Aiming to provide the basis for gene editing technology in the design and production of Corynebacterium glutamate strains and high value-added products.
