摘要
目的探究益母草碱对糖尿病脑病(DE)海马神经元细胞损伤及炎症的影响。方法利用高糖诱导的海马神经元(HT-22)细胞建立DE细胞模型,同时给予益母草碱干预,24 h后利用四氮唑盐比色法(MTT)检测海马神经元细胞损伤,利用酶联免疫吸附测定法(ELISA)检测细胞上清液中白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)水平,利用Western blot检测细胞中半胱氨酸天冬氨酸蛋白酶-3(caspase-3)、Bcl-2相关X蛋白(Bax)、B淋巴细胞瘤-2(Bcl-2)、脑源性神经营养因子(BDNF)蛋白表达情况。结果益母草碱能显著抑制高糖诱导的HT-22细胞活力,并降低IL-1β、TNF-α、IL-6水平。Western blot实验结果显示,益母草碱可以降低凋亡蛋白caspase-3、Bax的表达,促进抗凋亡蛋白Bcl-2的表达,提高高糖诱导的HT-22中BDNF的表达。结论益母草碱能通过抑制海马神经元凋亡及炎症反应来改善DE中高糖诱导的海马神经元损伤。
Objective To explore the effects of leonurine on hippocampal neuronal cells damage and inflammation in diabetic encephalopathy(DE).Methods Hippocampal neuron(HT-22)cells induced by high glucose were used to establish a DE cell model,and leonurine was administered at the same time.After 24 h,hippocampal neuronal cells damage was detected by methyl thiazolyl tetrazolium(MTT),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)levels in the supernatant of cells were detected by enzyme-linked immunoadsorbent assay(ELISA),and the protein expressions of cysteinyl aspartate specific proteinase-3(caspase-3),Bcl-2 associated protein(Bax),B-cell lymghoma(Bcl-2)and brain derived neurotrophic factor(BDNF)were detected by Western blot.Results After intervention of leonurine,the activity of HT-22 cells induced by high glucose was significantly inhibited and the levels of IL-1β,TNF-αand IL-6 decreased.Western blot results showed that leonurine could decrease the expressions of apoptotic protein caspase-3 and Bax,promote the expression of anti-apoptotic protein Bcl-2,and increase the expression of BDNF in HT-22 induced by high glucose.Conclusion Leonurine can improve hippocampal neuronal damage induced by high glucose in DE by inhibiting hippocampal neuronal apoptosis and inflammatory response.
