间隙连接蛋白家族在癌症中作用及其机制的研究进展

摘要 间隙连接蛋白(connexin,Cx)的研究始于上世纪五十年代,1957年Robertson通过电子显微镜观察其结构;1986年第一个Cx基因GJB1(编码Cx32蛋白)被成功克隆;2002年Willecke等对小鼠和人类基因组进行分析,对比其基因及编码蛋白的关系。编码Cx的基因家族包括多个成员,目前在人类和小鼠基因组中分别发现了21和20个基因。Cx根据其理论预测分子量来命名,如Cx31,表示分子量为31 kD的Cx。Cx广泛分布于机体的多种组织,发挥重要生理功能。其功能异常可导致多种疾病,包括先天性耳聋、先天性白内障、先天性心脏病、皮肤病、神经系统疾病、胃肠道和肝脏疾病、肺脏疾病和肿瘤等[1]。本文将重点综述Cx家族在肿瘤发生发展中的作用及其机制的研究进展。 Connexins are a family of transmembrane proteins with relatively conservative structure.Each member of connexins contains a similar monomer protein of four transmembrane domains.Connexins play critical roles in gap junctions,hemichannels,and protein-protein interaction.Studies suggest that connexin family can inhibit tumor growth or increase tumorigenicity by regulating the growth,migration and invasion of cancer cells.This review focuses on the research progress of connexin family in the pathogenesis of cancer.
