乳腺癌组织miR-411-3p、BPTF表达水平及临床意义研究 被引量:1

摘要 目的:探讨乳腺癌组织中微小RNA-411-3p(miR-411-3p)、溴区结构域转录因子(BPTF)表达与患者预后的关系。方法:选取2015年3月至2018年3月于本院手术治疗的96例女性乳腺癌患者的癌组织和72例乳腺癌旁正常组织为研究对象。采用实时荧光定量PCR法检测所有组织miR-411-3p、BPTF mRNA表达水平,免疫组化法检测BPTF表达。分析miR-411-3p、BPTF水平与乳腺癌患者临床病理特征的关系。Kaplan-Meier法分析miR-411-3p、BPTF水平与乳腺癌患者生存率的关系,多因素Cox回归分析影响乳腺癌预后因素。结果:Ualcan数据库中,乳腺癌组织中miR-411-3p表达水平低于癌旁组织(P<0.05)。与癌旁组织相比,乳腺癌组织中BPTF mRNA相对表达水平、BPTF阳性率较高(P<0.05),miR-411-3p相对表达水平较低(P<0.05),miR-411-3p、BPTF均与淋巴结转移、TNM分期相关(P<0.05)。Kaplan-Meier分析显示,miR-411-3p高表达组平均生存时间显著长于miR-411-3p低表达组(P<0.05);BPTF阴性平均生存时间显著长于BPTF阳性(P<0.05)。多因素分析结果显示,miR-411-3p低表达、BPTF阳性是乳腺癌患者不良预后的独立危险因素(P<0.05)。结论:miR-411-3p、BPTF与乳腺癌临床病理特征密切相关,miR-411-3p低表达、BPTF高表达是乳腺癌不良预后的危险因素。 Objective:To analyze the expression levels of microRNA-411-3p(miR-411-3p)and bromodomain PHD finger transcription factor(BPTF)in breast cancer tissues,and to explore their relationship with the prognosis of patients.Methods:A total of 96 cases of cancer tissues and 72 cases of normal tissues adjacent to breast cancer from breast cancer patients who were surgically treated in our hospital from March 2015 to March 2018 were selected as the research objects.Real-time fluorescence quantitative PCR was used to detect the expression levels of miR-411-3p and BPTF mRNA in all tissues,and the expression of BPTF in the tissues was detected by immunohistochemistry.The relationship between the levels of miR-411-3p and BPTF in breast cancer tissues and the clinicopathological characteristics of patients was analyzed,their relationship with the survival rate of breast cancer patients was analyzed with Kaplan-Meier method,and the factors affecting the prognosis of breast cancer was analyzed with multivariate Cox regression.Results:In the Ualcan database,the expression level of miR-411-3p in breast cancer tissues was lower than that in adjacent tissues(P<0.05).Compared with adjacent tissues,the relative expression level of BPTF mRNA and the positive rate of BPTF in breast cancer tissues were higher(P<0.05),the relative expression level of miR-411-3p was lower(P<0.05),miR-411-3p and BPTF were correlated with lymph node metastasis and TNM staging(P<0.05).Kaplan-Meier analysis showed that the average survival time of the miR-411-3p high expression group was significantly longer than that of the miR-411-3p low expression group(P<0.05);the average survival time of BPTF negative was significantly longer than that of BPTF positive(P<0.05).Multivariate analysis showed that low expression of miR-411-3p and positive BPTF were independent risk factors for poor prognosis in breast cancer patients(P<0.05).Conclusion:The expression of miR-411-3p is low and the expression of BPTF is high in breast tissue,they are closely related to the clinicopathological characteristics of breast cancer.Low expression of miR-411-3p and high expression of BPTF were risk factors for poor prognosis of breast cancer.
