沉香香粉熏香调节多神经递质途径的抗焦虑/抑郁作用研究 被引量:6

Study on the anti-anxiety/depression effect of agarwood fragrantpint incense inhalationvia regulating the multiple neurotransmitters balance

摘要 目的:探讨沉香香粉(AFP)加热熏香吸入对间氯苯哌嗪(MCPP)诱导的焦虑小鼠模型和慢性不可预知性温和刺激的抑郁小鼠模型的抑制作用并探究其作用机制。方法:通过评价旷场活动实验、明暗箱穿梭实验、悬尾实验、游泳实验等行为学变化,考察沉香香粉熏香吸入的抗焦虑和抗抑郁作用;采用ELISA法测定小鼠海马组织中神经递质五羟色胺(5-HT)、谷氨酸(Glu)和伽马氨基丁酸受体A(GABAA)的水平,同时利用Western blot检测沉香熏香对谷氨酸能系统代谢和转运相关蛋白谷氨酸受体1(GluR1)及囊泡谷氨酸转运蛋白1(VGluT1)蛋白表达的影响,探究沉香熏香吸入抗焦虑和抗抑郁的作用机制。结果:行为学数据显示,与焦虑模型比较,AFP熏香吸入组显著缩短运动总路程和降低运动速度,增加中央区域活动路程和运动速度,延长静止时间,降低动物在暗箱活动路径和速度;与抑郁模型比较,AFP组可显著缩短强迫游泳和悬尾不动时间;递质及受体调控方面:AFP组可显著降低焦虑模型组的Glu水平,增加GABAA、5-HT递质水平;显著升高抑郁模型组5-HT和Glu水平,降低GABAA,同时AFP组还显著升高焦虑和抑郁海马体中GluR1、VGluT1蛋白表达。结论:研究表明,沉香香粉熏香可显著抗焦虑/抑郁,其作用机制可能与调控神经递质分泌及其受体和转运蛋白的表达有关。 OBJECTIVE To explore the anti-anxiety and anti-depression of agarwood fragrantpint(AFP)heating incense and its mechanism in the anxiety mouse model induced by m-chlorophenylpiperazine(MCPP)and the depression mouse model induced by chronic unpredictable mild stimulation.METHODS The anti-anxiety and anti-depression effects of agarwood powder inhalation were investigated by evaluating the behavioral changes of open field activity test,light-dark chamber shuttle test,suspended tail test and swimming test.The levels of 5-hydroxytryptamine(5-HT),Glutamic acid(Glu)and gamma-aminobutyric acid A receptor(GABAA)levels were determined by ELISA in hippocampus.The protein expressions of the Glutamate receptor 1(GluR1)and Vesicular glutamate Transporter Type 1(VGluT1)was detected by Western Blot,which to explore the anti-anxiety and anti-depression mechanism of agarwood incense inhalation.RESULTS Behavioral data showed that AFP inhalation significantly reduced the total distance and speed of exercise,and increased the distance and speed of movement in the central region,increase the rest time,reducing the distance and speed in the dark box.Compared with depression model,AFP group significantly reduced the immobility time of TST and FST.In terms of neurotransmitter regulation,AFP group significantly reduced Glu level in anxiety model,increased the levels of GABAA,5-HT,increased the levels of 5-HT and Glu in depression model,decrease GABAA,and AFP group also significantly increased the protein expression of GluR1 and VGluT1 in hippocampus of anxiety and depression model mice.CONCLUSION Studies have shown that agarwood powder could significantly inhibit the anxiety/depression,and its mechanism may be related to the regulation the expressions of neurotransmitter and its receptor and transporter.
