摘要
目的 通过对激素性股骨头坏死(steroid-induced osteonecrosis of femoral head, SONFH)差异表达基因进行生物信息学分析,确定与SONFH的发生和发展相关的关键致病生物标志物,探究SONFH发生的分子机制。方法 从基因表达总表(GEO)数据库中获取SONFH患者与非SONFH患者外周血清中的基因表达谱,采用GEO2R在线工具软件分析,获得差异表达基因(DGEs)。通过String数据库构建SONFH差异表达基因蛋白质相互作用(PPI)网络,通过Cytoscape软件cytoHubba插件筛选出核心基因,并进行差异表达基因功能富集分析、通路富集分析和基因富集分析(GSEA)。结果 从GEO数据库中下载的基因表达序列GSE123568中共筛选出了719个SONFH差异表达基因,在各种关键途径中高度富集,包括破骨细胞分化、吞噬小体、趋化因子信号通路等。GSEA富集分析结果显示JAK-STAT信号通路、toll受体通路等与SONFH密切相关。结论 多个富集功能参与了SONFH发生的过程,研究还确定了10个关键基因,可能成为早期筛查的重要标志物。
Objective Bioinformatics analysis of the differentially expressed genes of steroid-induced osteonecrosis of femoral head(SONFH) was conducted to identify key pathogenic biomarkers related to the occurrence and development of SONFH and explore the molecular mechanism of SONFH occurrence. Methods Gene expression profiles of SONFH patients in peripheral serum and non-SONFH patients were obtained from the gene expression omnibus(GEO) database. GEO2R online tool software analysis was used to obtain differentially expressed genes(DGEs). SONFH differentially expressed gene protein interaction(PPI) network was constructed through String database, core genes were selected through Cytoscape software cytoHubba plugin, and functional enrichment analysis, pathway enrichment analysis and gene set enrichment analysis(GSEA) were performed. Results A total of 719 SONFH differentially expressed genes, selected from the gene expression sequence GSE123568 downloaded from the GEO database, were highly enriched in various key pathways, including osteoclast differentiation, phagosome, and chemokine signaling pathways. The results of the GSEA enrichment analysis showed that the JAK-STAT signaling pathway and the toll receptor pathway are closely related to the SONFH. Conclusion Multiple enrichment functions were involved in the process of SONFH occurrence, and the studies identified 10 key genes that may be important markers for early screening.
