摘要
探究分心木水提物对大鼠慢性非细菌性前列腺炎的作用.通过脂多糖(LPS)诱导RAW264.7巨噬细胞炎症模型,发现分心木水提物对LPS诱导巨噬细胞释放NO的抑制作用最好.利用角叉菜胶建立大鼠慢性非细菌性前列腺炎(CNP)模型,发现分心木水提物能在一定程度上减轻大鼠前列腺组织水肿及炎性细胞浸润,降低CNP大鼠血清中前列腺特异性抗原(PSA)、肿瘤坏死因子-α(TNF-α)、前列腺素E_(2)(PGE_(2))和前列腺组织中丙二醛(MDA)的含量,降低前列腺组织中环氧合酶2(COX-2)的基因表达水平,提升前列腺组织中NF-E_(2)相关因子2(Nrf_(2))的基因表达水平.实验结果表明:分心木水提物可能通过COX-2/PGE_(2)和Nrf2两条通路、多成分协同作用的方式抑制炎症因子的生成以及减轻氧化应激反应,进而对CNP大鼠起到治疗作用.
The aim of this study is to investigate the effects of Diaphragma Juglandis Fructus(DJF)water extract on Chronic Non-bacterial Prostatitis(CNP)in rats.Using the Lipopolysacchaide(LPS)-induced infl amma-tion model in RAW264.7 macrophages,it was found that DJF water extract exhibited the strongest inhibitory effect on LPS-induced NO release from macrophages.A rat model of CNP was established using carrageenan.The results showed that DJF water extract could alleviate the edema and inflammatory cell infiltration in the pros-tate tssue of CNP rats to a certain extent.It also reduced the levels of Prostate-Specific Antigen(PSA),Tumor Necrosis Factor-α(TNF-α),and Prostaglandin E_(2)(PGE_(2))in serum,as well as Malondialdehyde(MDA)content in prostate tissue.Futhermore,the extract decreased the gene expression level of Cyclooxygen ase2(COX-2)and increased the expression of NF-E_(2)related factor2(Nrf2)in prostate tsse.Based on the experimental results,DJF water extract may inhibit the production of inflammatory factors and reduce oxidative stress reactions through COX-2/PGE_(2)and Nrf2 pathways,as well as by employing multi-component syn-ergistic action,demonstrating therapeutic effects on CNP in rats.
