同源域蛋白同源盒在胃癌组织的表达及其临床意义 被引量:1

Expression and clinical significance of homeodomain-only protein homeobox in gastric cancer

摘要 目的探讨同源域蛋白同源盒(HOPX)在胃癌中的表达、预后预测价值及临床意义。方法通过癌症基因组图谱(TCGA)数据库下载胃癌转录组数据及临床相关参数,利用R语言分析基因HOPX的表达差异及临床病理特征的相关性。通过基因表达交互网站(GEPIA)及生存分析数据库(K-M)及多因素回归分析胃癌患者预后影响因素。同时,通过R语言分析基因HOPX的相关通路,探索其在胃癌中的潜在机制。结果HOPX在胃癌组织中的mRNA表达水平显著高于正常组织[2.929(2.042,3.805)比1.963(1.500,3.040),Z=15.5,P<0.05],其表达量与肿瘤分化程度、肿瘤浸润深度、TNM分期呈正相关(P<0.05);TCGA数据库及K-M plotter数据库分析显示胃癌组织中HOPX高表达患者预后差(P<0.05);多因素分析结果显示HOPX表达[风险比=1.595,95%可信区间:1.087~2.340,P<0.05]是影响胃癌预后的独立因素;KEGG通路富集分析显示HOPX参与了细胞黏附通路、细胞外基质受体相互作用通路、蛋白质消化与吸收通路、血管平滑肌收缩通路、PI3K-Akt信号通路等相关通路。结论HOPX mRNA在胃癌组织中高表达,且与胃癌患者的不良预后密切相关,可作为胃癌预后的独立危险因子,其可能通过细胞黏附通路、PI3K-Akt信号通路等在胃癌中发挥作用。 Objective To explore the expression,prognostic value and clinical significance of homeodomain-only protein homeobox(HOPX)in gastric cancer(GC).Methods The transcriptome data and clinical parameter of GC were downloaded through the Cancer Genome Atlas(TCGA)database.The mRNA expression of HOPX and its correlation with clinicopathological features were analyzed by R.The prognostic factors of GC patients were explored by the Gene Expression Profiling Interactive Analysis(GEPIA)database,the Kaplan-Meier plotter(K-M plotter)database and Multivariate analysis.At the same time,the relevant pathways of HOPX were analyzed by R in order to explore its potential mechanism in GC.Results The mRNA expression of HOPX in GC was significantly higher[2.929(2.042,3.805)vs.1.963(1.500,3.040),Z=15.5,P<0.05],and its expression was significantly correlated with the tumor differentiation degree,tumor invasion depth,TNM stage(P<0.05).TCGA database and K-M plotter database analysis showed that the overall survival time/progression-free survival time of GC patients with high HOPX expression were shorter than that with low HOPX expression(P<0.05).Moreover,Multivariate analysis showed that HOPX expression[hazard ratio=1.595,95%confidence interval:1.087-2.340,P<0.05]was an independent predictor of the prognosis of GC patients.The KEGG pathway enrichment analysis showed that HOPX-related differential genes were mainly enriched in the cell adhesion pathway,extracellular matrix receptor interaction pathway,protein digestion and absorption pathway,vascular smooth muscle contraction pathway,PI3K-Akt signaling pathway and other related pathways.Conclusion HOPX mRNA is highly expressed in GC and closely related to the poor prognosis of GC patients.The expression of HOPX is an independent risk factor for GC.Moreover,it may play an important role in GC through cell adhesion pathway and PI3K-Akt signaling pathway.
