基于独立成分分析的睡眠剥夺静息态默认模式网络研究 被引量:2

A Study on the Change of Resting State Functional Connection of Default Mode Network in Sleep Deprivation Based on Independent Component Analysis

摘要 目的使用独立成分分析方法探讨睡眠剥夺与正常睡眠后默认模式网络(DMN)功能连接的差异。方法招募50例健康志愿者分别于24 h剥夺睡眠后与正常睡眠后采集静息态功能磁共振图像,同时使用精神运动警觉性任务(PVT)收集持续性注意力行为学数据。采用独立成分分析方法(ICA)提取静息状态下剥夺睡眠后与正常睡眠后的DMN成分,应用配对样本t检验(P<0.05,FDR校正)比较两种不同睡眠状态下DMN的功能活动强度差异。同时使用Pearson相关分析探讨差异脑区的活动强度与PVT脱漏频次的相关性。结果与正常睡眠状态相比,睡眠剥夺后左侧内侧前额叶(X,Y,Z=-6,53,4,t=3.89)、右侧内侧前额叶(X,Y,Z=7,59,-4,t=4.05)、左侧楔前叶(X,Y,Z=-2,-60,31,t=4.73)及右侧后扣带回(X,Y,Z=2,-49,30,t=4.27)的功能连接均显著增加(FDR校正,P<0.05,团块大小≥20体素),且发现右侧后扣带回的功能连接强度差值与PVT脱漏频次呈显著正相关(r=0.717,P<0.001)。结论DMN相关脑区的异常激活可能是睡眠剥夺后持续性注意力下降的重要神经机制。 Objective To investigate the difference of functional connectivity of brain default mode network(DMN)between sleep deprivation and rest wakefulness by using independent component analysis(ICA).Methods Fifty healthy subjects were recruited to collect resting state fMRI images after rest wakefulness and after 24 hours of sleep deprivation respectively.Psychomotor vigilance test(PVT)task was used to measure sustained attention.The ICA was used to extract DMN component of resting-state fMRI after rest wakefulness and after 24 hours of sleep deprivation separately.The differences of functional connectivity between sleep deprivation and rest wakefulness were compared by paired sample t test(P<0.05,FDR corrected).Pearson correlation analysis was used to investigate the relationships between the altered functional connectivity of DMN and PVT task performance.Results The DMN data showed that compared with after rest wakefulness,the functional connectivity of the left medial prefrontal cortex(X,Y,Z=-6,53,4,t=3.89),on the right medial prefrontal cortex(X,Y,Z=7,59,-4,t=4.05),the left precuneus(X,Y,Z=-2,-60,31,t=4.73)and the right posterior cingulate gyrus(X,Y,Z=2,-49,30,t=4.27)was significantly higher after sleep deprivation(FDR corrected,P<0.01,cluster size≥20 voxel).It was noteworthy that the functional connectivity of the right posterior cingulate gyrus was positively correlated with PVT task performance(r=0.717,P<0.001).Conclusion Abnormal activation of DMN related brain regions may underlie the sustained attention deficits after sleep deprivation.
