Caveolin-1蛋白和妊娠期糖尿病产妇胎盘组织中糖脂代谢信号转导通路和妊娠结局的关联研究 被引量:1

Study on the association between Caveolin-1 protein and glycolipid metabolism signal transduction pathway in placental tissues of pregnant women with gestational diabetes mellitus and pregnancy outcome

摘要 目的 探讨小凹蛋白-1(Cav-1)与妊娠期糖尿病(GDM)产妇胎盘组织中糖脂代谢信号转导通路和妊娠结局的关系。方法 选择2021年1—12月于本院进行产检并住院分娩的GDM产妇所产新生儿120例作为研究对象,根据产妇分娩结果将其分为GDM合并巨大儿(GDMM组,46例)与GDM合并正常新生儿(GDMN组,74例),另选同期在本院产检并住院分娩的正常产妇所产正常新生儿60名作为正常对照组。检测GDM产妇胎盘组织中Cav-1、GLUT1、p-AMPK(Thr172)以及p-ACC(Ser79)的蛋白表达水平并进行分析。结果 GDMM组产妇体质指数(孕前、分娩前)、孕期体重增加、空腹血糖(孕中期、孕晚期)、新生儿出生体重均分别明显高于GDMN组(P<0.05)。GDMM组产妇Cav-1、p-AMPK(Thr172)及p-ACC(Ser79)蛋白表达量均分别明显低于GDMN组,GLUT1蛋白表达量明显高于GDMN组(P<0.05)。新生儿出生体重与胎盘组织Cav-1、p-AMPK(Thr172)及p-ACC(Ser79)蛋白表达呈负相关(均P<0.05),与胎盘组织GLUT1呈正相关(P<0.05);胎盘组织Cav-1与GLUT1蛋白表达呈负相关(P<0.05),与p-AMPK(Thr172)及p-ACC(Ser79)蛋白表达呈正相关(均P<0.05);胎盘组织GLUT1与p-AMPK(Thr172)及p-ACC(Ser79)蛋白表达呈负相关(均P<0.05);胎盘组织p-AMPK(Thr172)与p-ACC(Ser79)蛋白表达呈正相关(P<0.05)。结论 GDM产妇胎盘组织中糖脂代谢信号转导通路相关蛋白的表达与新生儿出生体重密切有关,并参与了巨大儿的发生,应予以重视。 Objective To discuss the relationship between caveolin-1(Cav-1)and the signal transduction pathway of glycolipid metabolism in placenta tissues and the pregnancy outcomes of gestational diabetes mellitus(GDM)women.Methods 120 newborns birthed by GDM mothers those received pregnancy check-up and give birth in our hospital form Jan 2021 to Dec 2021 were selected as research objects.According to the pregnancy outcome,they were divided into the GDM complicated with macrosomia group(GDMM group,46 cases)and the GDM complicated with normal infant group(GDMN group,74 cases),another 60 cases of normal newborn birthed by normal pregnancy women in our hospital during the same period were selected as the normal control group.The protein expression level of Cav-1,GLUT1,p-AMPK(thr172)and p-ACC(ser79)in placenta of GDM women were detected and analyzed.Results The body mass index(before pregnancy,before delivery),weight gain during pregnancy,fasting blood glucose(the second and third trimester)and birth weight of newborn of the GDMM group were significantly higher than the GDMN group(P<0.05).The expression of Cav-1,p-AMPK(Thr172)and p-ACC(Ser79)protein of the GDMM group were significantly lower than the GDMN group,however,the expression of GLUT1 protein was significantly higher than the GDMN group(P<0.05).The newborn birth weight was negatively correlated with the expression of Cav-1,p-AMPK(Thr172)and p-ACC(Ser79)protein in placenta(all P<0.05),and positively correlated with the expression of GLUT1 in placenta(P<0.05).The expression of Cav-1 protein in placenta is negatively correlated with GLUT1(P<0.05),and positively correlated with the expression of p-AMPK(Thr172)and p-ACC(Ser79)in placenta(all P<0.05).The expression of GLUT1 protein in placenta is negatively correlated with p-AMPK(Thr172)and p-ACC(Ser79)(all P<0.05).The expression of p-AMPK(Thr172)protein in placenta is positively correlated with p-ACC(Ser79)(P<0.05).Conclusions The expression of glycolipid metabolism signal transduction pathway related proteins in placentas of prenancy women with GDM is closely related to the birth weight of newborn,and which is involved in the development of macrosomia,so we should pay attention to it.
