肝癌患者血清中lncRNA HOTAIR表达与临床病理特征的相关性及诊断价值

Correlation Between the Expression of lncRNA HOTAIR in Serum of Patients with Hepatocellular Carcinoma and Clinicopathological Characteristics and Its Diagnostic Value

目的分析血清长链非编码RNA HOX转录反义RNA(IncRNA HOTAIR)在肝癌患者中的表达与临床诊断意义。方法选取100例肝癌患者为研究对象,并将其纳入肝癌组,另选取同期收治的80例肝硬化患者纳入肝硬化组,同时选取同期行体检的75例健康体检者纳入对照组。采集3组5 ml静脉血,以实时荧光定量逆转录聚合酶链反应(RT-PCR)检测对比3组血清IncRNA HOTAIR相对表达量。同时,收集肝癌患者的年龄、性别等一般资料,分析血清IncRNA HOTAIR与其临床病理特征的关系;另外绘制受试者工作曲线(ROC),分析血清IncRNA HOTAIR在肝癌中的诊断效能。结果肝癌组的IncRNA HOTAIR相对表达量[(2.53±0.58)]高于肝硬化组[(0.81±0.25)]与对照组[(0.20±0.05)],差异有统计学意义(P<0.05)。IncRNA HOTAIR表达与肝癌患者的TNM分期、血管侵犯、肝外转移有关,差异有统计学意义(P<0.05);IncRNA HOTAIR表达与患者的年龄、性别、肿瘤数目无关,差异无统计学意义(P>0.05)。ROC结果显示:血清IncRNA HOTAIR诊断肝癌的AUC为0.901(95%CI:0.852~0.949),具有较高的诊断价值。结论肝癌患者血清内IncRNA HOTAIR呈高表达,其表达水平与患者TNM分期、血管侵犯、肝外转移相关,可能参与肝癌的发生、发展,在肝癌中具有较高的临床诊断意义。

Objective To analyze the expression of serum long chain non-coding RNA HOX transcriptional antisense RNA(IncRNA HOTAIR)in patients with hepatocellular carcinoma and its clinical diagnostic significance.Methods 100 patients with liver cancer were selected as the study object and included in the liver cancer group,another 80 patients with liver cirrhosis admitted in the same period were included in the liver cirrhosis group,and 75 healthy people who underwent physical examination in the same period were included in the control group.Collect 5 ml of venous blood from 3 groups,and measure the relative expression of serum InCRNA HOTAIR in three groups by real-time fluorescence quantitative reverse transcriptase polymerase chain reaction(RT-PCR).At the same time,general data such as age and sex of patients with liver cancer were collected to analyze the relationship between serum InCRNA HOTAIR and its clinicopathological characteristics;In addition,the subject working curve(ROC)was drawn to analyze the diagnostic efficacy of serum InCRNA HOTAIR in liver cancer.Results The relative expression of IncRNA HOTAIR in liver cancer group[(2.53±0.58)]was higher than that in liver cirrhosis group[(0.81±0.25)]and control group[(0.20±0.05)],the difference was statistically significant(P<0.05);The expression of IncRNA HOTAIR was correlated with TNM stage,vascular invasion and extrahepatic metastasis of HCC patients,and the difference was statistically significant(P<0.05);The expression of IncRNA HOTAIR was not related to the age,sex and tumor number of patients,and the difference was not statistically significant(P>0.05).The ROC results showed that the AUC of serum IncRNA HOTAIR in the diagnosis of liver cancer was 0.901(95%CI:0.852~0.949),which has high diagnostic value.Conclusion The expression level of IncRNA HOTAIR in the serum of patients with hepatocellular carcinoma is highly correlated with TNM stage,vascular invasion and extrahepatic metastasis.It may be involved in the occurrence and development of hepatocellular carcinoma and has high clinical diagnostic significance in hepatocellular carcinoma.
