直肠癌组织中ADAM12、A2aR表达水平与神经脉管侵犯及5-FU化疗敏感性的关系

Correlation of ADAM12 and A2aR Expression with Neurovascular Invasion and Sensitivity to 5-FU Chemotherapy in Patients with Rectal Cancer

目的探究直肠癌组织中ADAM12、A2aR表达水平与神经脉管侵犯及5-FU化疗敏感性的关系。方法选取62例新型辅助放化疗治疗前活检石蜡标本,采用免疫组化法检测直肠癌组织中ADAM12、A2aR表达水平,采用直肠癌消退分级标准对直肠癌组织化疗敏感性进行评价。结果A2aR蛋白的表达与肿瘤大小、患者性别、年龄大小、神经脉管侵犯程度无关(P>0.05),与TNM分期、浸润程度有关(P<0.05)。ADAM12蛋白的表达与肿瘤大小、患者性别、年龄大小、神经脉管侵犯程度无关(P>0.05),与浸润程度、TNM分期有关(P<0.05)。直肠癌化疗敏感性与浸润程度、TNM分期、A2aR和ADAM12表达强度有关(P<0.05)。直肠癌组织中A2aR表达与5-FU化疗敏感性呈正相关(γ=0.352,P<0.001)。直肠癌组织中ADAM12表达与5-FU化疗敏感性呈正相关(γ=0.276,P<0.030)。结论直肠癌组织中ADAM12、A2aR阳性表达率较高,对神经脉管侵犯度不高,与5-FU化疗敏感性呈现正相关。

Objective To investigate the correlation of Meltrinα(ADAM12)and adenosine A2a receptor(A2aR)expression with neurovascular invasion and sensitivity to 5-fluorouracil(5-FU)chemotherapy in patients with rectal cancer.Methods The pre-treatment biopsy paraffin specimens of 62 patients with novel adjuvant radiotherapy were collected,and immunohistochemical method was used to detect the expression levels of ADAM12 and A2aR in rectal cancer tissues.The radiochemotherapy sensitivity was evaluated based on rectal cancer regression grade(RCRG).Results The expression of A2aR protein was not correlated with tumor size,gender,age or neurovascular invasion degree(all P>0.05),while was correlated with TNM stage and infiltration degree(P<0.05).ADAM12 protein expression was not associated with tumor size,gender,age,or degree of neurovascular invasion(all P>0.05),while was associated with degree of infiltration and TNM stage(P<0.05).The sensitivity of chemoradiotherapy was related to the infiltration degree,TNM stage,and the expression intensity of A2aR and ADAM12 in rectal cancer patients(all P<0.05).A2aR expression in rectal cancer tissues was positively correlated with the 5-FU sensitivity(γ=0.352,P<0.001).ADAM12 expression in rectal cancer tissues was also positively correlated with the sensitivity of 5-FU sensitivity(γ=0.276,P<0.030).Conclusion The rectal cancer patients have an increased positive expression rate of ADAM12 and A2aR and not severe neurovascular invasion,furthermore,the expression of 2 indicators has a positive correlation with the 5-FU sensitivity.
