三七总皂苷对缺血性脑卒中肠道功能影响的探讨

摘要 目的探讨三七总皂苷对缺血性脑卒中后肠道相关蛋白及肠道功能的影响。方法将36只SPF级SD大鼠随机分为3组:假手术组、模型组和治疗组,3组继续分为7 d和14 d两个亚组。通过苏木素和伊红染色观察肠道病理学变化;采用Chiu病理评分评估肠道的组织学改变程度;通过免疫组织化学检测肠道Occludin和ZO-1蛋白的表达量。结果在假手术组中,肠黏膜正常,绒毛排列整齐,上皮完整;模型组肠道绒毛数量减少、形态破坏,治疗组与模型组的Chiu病理评分相比,差异具有统计学意义(P<0.05)。模型组肠道的Occludin和ZO-1蛋白表达量减少,治疗后肠道Occludin和ZO-1蛋白表达量有所恢复,在不同时间点,治疗组的Occludin蛋白表达量与模型组比较,差异具有统计学意义(P<0.05);在不同时间点,治疗组的ZO-1蛋白表达量与模型组比较,差异具有统计学意义(P<0.05)。结论三七总皂苷在治疗缺血性脑卒中时可以通过改善肠道的Occludin和ZO-1蛋白表达量,改善肠道功能,从而达到治疗目的。 Objective To investigate the effects of panax notoginseng saponins on intestinal related proteins and intestinal function of patients after ischemic stroke.Methods Thirty-six specific-pathongen-free(SPF)SD rats were randomly divided into three groups:the sham operation group,the model group and the treatment group.The three groups were further divided into 7-day and 14-day subgroups.Intestinal pathological changes were observed by hematoxylin and eosin staining.Chiu pathological scoring was used to evaluate the degree of intestinal histological changes.The expressions of intestinal Occludin and ZO-1 protein were detected by immunohistochemical assay.Results In the sham operation group,the intestinal mucosa was normal,the villi were neatly arranged,and the epithelium was intact.In the model group,the number of intestinal villi decreased and their morphology was damaged.The difference in Chiu pathological scoring between the treatment group and the model group was statistically significant(P<0.05).The protein expressions of intestinal Occludin and ZO-1 decreased in the model group,and they recovered a bit after treatment.At different time points,the difference in Occludin expression between the treatment group and the model group was statistically significant(P<0.05),and the difference in ZO-1 protein expression between the treatment group and the model group was statistically significant(P<0.05).Conclusion Panax notoginseng saponins can improve intestinal function by regulating protein expressions of intestinal Occludin and ZO-1 in the treatment of ischemic stroke.
