摘要
为探究肌醇代谢调控因子IolR对嗜水气单胞菌NJ-35的生存及环境应激的调控作用,检测了野生株、iolR缺失株(ΔiolR)及其互补株(CΔiolR)对不同碳源的代谢能力、耐酸、耐药、耐氧化、耐高渗以及抗小鼠单核/巨噬细胞RAW264.7吞噬能力,同时检测其对巨噬细胞的毒性作用。结果显示,ΔiolR缺失株对葡萄糖、果糖、甘露糖和半乳糖等4种碳源的代谢能力均显著降低,菌株的耐酸、耐氧化能力显著减弱,而耐高渗能力略有增强;巨噬细胞对ΔiolR的吞噬率升高63.33%。另外,药敏试验结果显示,ΔiolR缺失株对庆大霉素、妥布霉素、头孢曲松、头孢噻肟等4种抗生素的敏感性显著增强。研究表明,肌醇代谢调控因子IolR在嗜水气单胞菌的生存以及对不同胁迫环境的适应性方面发挥重要的调控作用。
In order to explore the regulation effect of inositol metabolism regulator IolR on the survival and environmental stress response of Aeromonas hydrophophilus NJ-35,the metabolic ability of wild strains,ΔiolR and CΔiolR,their acid resistance,drug resistance,oxygen resistance,hyperosmosis resistance in response to different carbon sources,were tested,and their anti-phagocytosis against mouse monocyte/macrophage RAW264.7 was also tested.At the same time,their toxicity to macrophages was detected.The results showed that the metabolic ability ofΔiolR against glucose,fructose,mannose and galactose was significantly reduced.Their acid resistance and oxidation resistance againstΔiolR was also significantly reduced,but their hyperosmotic resistance was enhanced slightly.The phagocytosis rate of ΔiolR by macrophages increased by 63.33%.In addition,the sensitivity test results showed that the sensitivity of the ΔiolR strain to gentamicin,tobramycin,ceftriaxone and cefotaxime was significantly enhanced.This study indicated that IolR,a regulator of inositol metabolism,played a role in regulating the survival and adaptability of Aeromonas hydrophila in response to different stress environments.
