ZEA、DON及其联合作用对李斯特菌感染小鼠血清免疫相关细胞因子的影响

Effects of ZEA,DON and their combined effects on serum immune-related cytokines in Listeria infected mice

摘要 旨在探究玉米赤霉烯酮(ZEA)、脱氧雪腐镰刀菌烯醇(DON)及其联合作用对细菌感染动物防御能力的影响。试验以小鼠为对象,研究了低剂量的ZEA、DON对李斯特菌感染小鼠血清中免疫相关因子的变化。试验分别设空白对照组(Con)、李斯特菌组(Lis)、10 mg/kg ZEA+李斯特菌组(ZEA+Lis)、1 mg/kg DON+李斯特菌组(DON+Lis)、10 mg/kg ZEA+1 mg/kg DON+李斯特菌组(ZEA+DON+Lis),染毒方式为灌胃,细菌感染方式为尾静脉注射,剂量5×104CFU。在试验结束后,用抗体芯片技术检测小鼠血清中免疫相关细胞因子的水平。结果显示,与Con组相比,Lis组免疫细胞因子如白介素IL-1β、IL-12P40/P70,集落刺激因子MCSF,趋化因子MIG皆显著升高(P<0.05)。与Lis组相比,ZEA+Lis组IL-1β显著下降(P<0.05),基质金属蛋白酶TIMP-1显著上升(P<0.05);DON+Lis组IL-1β、IL-12P40/P70和MCSF显著下降(P<0.05),趋化因子BLC TIMP-1显著或极显著上升(P<0.05或P<0.01);ZEA+DON+Lis组IL-1β与IL-12P40/P70显著下降(P<0.05),集落刺激因子GCSF、趋化因子BLC与MIG显著或极显著上升(P<0.05或P<0.01)。与ZEA组相比,ZEA+DON+Lis组IL-6、趋化因子MIP-1α与MIG、TIMP-1显著上升(P<0.05),趋化因子LIX显著下降(P<0.05)。与DON+Lis组相比,ZEA+DON+Lis组GCSF与MCSF显著或极显著上升(P<0.05或P<0.01),MIP-1α显著上升(P<0.05)。结果表明,低剂量的ZEA、DON及其联合均能不同程度地降低李斯特菌感染小鼠血清中免疫相关细胞因子的水平,说明ZEA、DON及其联合作用下机体对李斯特菌感染的防御能力降低;但在经剂量下,ZEA和DON联合暴露对小鼠血清中免疫相关细胞因子表达表现出具有一定的拮抗作用。 In order to explore the changes in the defensive ability of animals against bacterial infection under the background of zearalenone(ZEA),deoxynivalenol(DON)and their combined effects,mice were used as subjects of research here,and low-dose changes in immunerelated factors in the serum of mice infected with Listeria by ZEA and DON were studied.In the experiment,the blank control group(Con),the Listeria group(Lis),the ZEA(10 mg/kg)+Listeria group(ZEA+Lis),the DON(1 mg/kg)+Listeria group(DON+Lis),and the ZEA(10 mg/kg)+DON(1 mg/kg)+Listeria group(ZEA+DON+Lis)were formed.The method of exposure was gavage,and the method of bacterial infection was tail vein injection.The levels of immune-related cytokines in the serum of the mice were detected by antibody chip technology after the test.The results showed that,compared with the Con group,the immune cytokines such as interleukin IL-1β,IL-12P40/P70,the colony stimulating factor MCSF,and the chemokine MIG were significantly increased in the Lis group(P<0.05).Compared with the Listeria group,the interleukin IL-1β in the ZEA+Lis group was significantly decreased(P<0.05),and the matrix metalloproteinase TIMP-1 was significantly increased(P<0.05).The colony factor MCSF decreased significantly(P<0.05),the chemokine BLC and the matrix metalloproteinase TIMP-1 increased significantly or extremely significantly(P<0.05 or P<0.01).In the ZEA+DON+Lis group,interleukin IL-1β and IL-12P40/P70 decreased significantly(P<0.05),and the colony stimulating factor GCSF,chemokine BLC and MIG increased significantly(P<0.05 or P<0.01).Compared with the ZEA group,the interleukin IL-6,the chemokines MIG and MIP-1α,and the matrix metalloproteinase TIMP-1 in the ZEA+DON+Lis group were significantly increased(P<0.05),and their LIX was significantly decreased(P<0.05).Compared with the DON+Lis group,the colony-stimulating factors GCSF and MCSF of the ZEA+DON+Lis group were significantly or extremely significantly increased(P<0.05 or P<0.01),and their chemokine MIP-1αwas significantly increased(P<0.05).These results indicated that low doses of ZEA,DON and their combination reduced the levels of immune-related cytokines in the serum of Listeria-infected mice to varying degrees,suggesting the body defended itself against Listeria infection under the action of ZEA,DON and their combination.However,at the test dose,the combined exposure of ZEA and DON showed a certain antagonistic effect on the expression of immune-related cytokines in the serum of the mice.
