摘要
探讨不同浓度的桑黄水提物对U14荷瘤小鼠的肿瘤抑制作用及对肠道菌群的影响。昆明小鼠随机分为正常对照组、模型组、桑黄水提物(SHWE)低剂量组、SHWE中剂量组、SHWE高剂量组,每组各12只。皮下移植构建U14宫颈癌荷瘤小鼠模型,SHWE组小鼠灌胃不同剂SHWE,1次/d,连续给药21 d。末次给药24 h后观察记录各组小鼠的体质量、检测肿瘤组织抑瘤率、肿瘤体积变化,计算生存率。采集小鼠肠道内容物,通过16srDNA扩增子测定肠道菌群含量。结果显示:SHWE具有很好的抗肿瘤作用,SHWE高、中、低剂量组的抑癌率分别为44.26%、67.54%和62.30%,并且毒副作用小,在中剂量的SHWE干预下小鼠整体状态更好,死亡率低,小鼠体质量和体重变化和空白组趋于一致;在SHWE的干预作用下,总的来看小鼠肠道菌群的丰度和多样性要高于模型组。研究显示,在U14宫颈癌荷瘤小鼠模型中,SHWE具有很好的抗肿瘤作用并能通过改变肠道菌群的构成来影响肿瘤的发展。
To investigate the effect of different concentrations of water extract of Sanghuang on tumor inhibition and intestinal flora in U14 tumor-bearing mice.Methods:Kunming mice were randomly divided into normal control group,model group,low dose group,middle dose group and high dose group,12 mice in each group.U14 cervical cancer tumor-bearing mice model was established by subcutaneous transplantation.Mice in water extract of Sanghuang group were treated with intragastric administration of different doses of water extract of Sanghuang once a day for 21 days.After 24 h of the last administration,the body weight of mice in each group was observed,the tumor inhibition rate and tumor volume were detected,and the survival rate was calculated.The intestinal contents of mice were collected and the content of intestinal flora was determined by 16 s rDNAamplicon.Results:The water extract of Phellinus igniarius had a good anti-tumor effect.The tumor inhibition rates of the high,medium and low dose groups were 44.26%,67.54%and 62.30%,respectively,and the toxic and side effects were small.Under the intervention of the medium dose of the water extract of Phellinus igniarius,the overall state of the mice was better,the mortality rate was low,and the and body weight of the mice were consistent with those of the blank group.Under the intervention of aqueous extract of Sanghuang,the abundance and diversity of intestinal flora in mice were higher than those in model group.Conclusion:In the U14 cervical cancer tumor-bearing mouse model,Sanghuang aqueous extract has a good anti-tumor effect and can affect the development of tumors by changing the composition of intestinal flora.
