黄芪多糖对奶山羊精液冷冻保存效果的影响 被引量:3

摘要 为了切实提高奶山羊精液冷冻保存效果,并为新型精液冷冻稀释液配方的研发提供试验依据,该研究在奶山羊精液冷冻稀释液中添加不同浓度的黄芪多糖,冷冻-解冻后检测精子活力、运动参数、抗氧化性能、线粒体膜电位等指标。结果表明:在奶山羊精液冷冻稀释液中添加0.2 g/L黄芪多糖能够显著提高精液冷冻保存效果(精子活力、质膜完整率、顶体完整率分别达到52.89%、55.34%、50.31%,直线运动速率和曲线运动速率分别达到45.84μm/s和59.46μm/s),提高精子冻后总抗氧化能力(total antioxidant capacity,T-AOC)、超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalse,CAT)的活性,降低精子内活性氧(Reactive oxygen species,ROS)水平和氧化产物丙二醛(malondialdehyde,MDA)的含量,提高冷冻-解冻后精子线粒体膜电位和ATP水平,从而有效改善精液冷冻保存效果。研究表明,在奶山羊精液冷冻稀释液中添加0.2 g/L黄芪多糖能够减少奶山羊精子冷冻过程中的氧化损伤,提高奶山羊精液冷冻保存质量,为奶山羊精液冷冻保存和其在人工授精方面的应用提供科学理论参考。 To improve the cryopreservation effect of dairy goat semen and provide a scientific theoretical basis for a new type of formulation of semen freezing diluent,different concentrations of astragalus polysaccharides were added to the frozen diluent of dairy goat semen,and the sperm motility,movement parameters,antioxidant properties and mitochondrial membrane potential were detected after freezing-thawing.The results showed that 0.2 g/L astragalus polysaccharide significantly increased the viability(52.89%),acrosome integrity(50.31%),plasma membrane integrity(55.34%),total antioxidant capacity(T-AOC)and the antioxidant enzyme SOD and CAT activities of frozen-thawed sperm,linear and curvilinear velocities reached 45.84μm/s and 59.46μm/s,respectively,while it significantly reduced the ROS level and malondialdehyde(MDA)content(P<0:05).In addition,further analysis showed that 0.2 g/L APS significantly increased the mitochondrial membrane potential and ATP levels of frozen-thawed sperm,and thus improved the cryopreservation effect of semen effectively.In summary,0.2 g/L astragalus polysaccharide protects dairy goat semen from reducing the oxidative damage during cryopreservation,and improve the cyopreservation quality of dairy goat semen.It provides a scientific and theoretical reference for the cyopreservation and application of frozen semen in artificial insemination.
