基于UPLC-ESI-Q-TOF-MS/MS技术的复方钩藤降压片化学成分分析 被引量:3

Study on Chemical Constituents from Compound Uncaria Antihypertensive Tablets Based on UPLC-ESIQ-TOF-MS/MS

摘要 目的 采用液质联用技术对复方钩藤降压片的化学成分进行快速分析与鉴定。方法 应用超高效液相色谱-电喷雾四极杆-飞行时间质谱(UPLC-ESI-Q-TOF-MS/MS)对复方钩藤降压片中主要的化学成分进行系统分析,采用Agilent Masshunter Qualitative Analysis工作站中的分子特征提取(MFE)功能和中药成分数据库(TCM-DATA),根据正负离子模式质谱扫描提供化合物一级、二级质谱数据的质谱碎片离子信息进行定性分析,同时对复方钩藤降压片中的各类成分进行鉴定,并通过单味药材的比对分析归属其化学成分的中药来源。结果 通过液质分析从复方钩藤片中共鉴定出72个化学成分,其中生物碱类化学成分30个,黄酮及其苷类化学成分10个、单萜类化合物6个,酚酸及其苷类化学成分14个,甾体及有机酸类等化学成分12个。结论采用UPLC-ESI-Q-TOF-MS/MS技术可对复方钩藤降压片进行快速和系统的化学成分识别,为其质量控制和临床应用提供了理论依据。 Objective To clarify the chemical constituents from Uncaria Antihypertensive Tablets and establish an effective LC-MS method for the identification of complex composition.Methods UPLC-ESI-Q-TOF-MS/MS was used to analyze the chemical constituents of Uncaria Antihypertensive Tablets. The analysis was performed by molecular feature extraction(MFE) and the Chinese herbal ingredients database(TCM-DATA) in Agilent Masshunter Qualitative Analysis station. We conducted qualitative analysis of the chemical constituents in Uncaria Antihypertensive Tablets depending on the information of the first and second order accurate mass and the multistage fragment ions data from spectrometry scanning. Meanwhile various components in Uncaria Antihypertensive Tablets were identified. Components were assigned to the traditional Chinese medicine through comparative analysis of its single herbs.Results A total of 72 compounds were identified in this study,including 30 alkaloids,10 flavonoids and their glycosides, 6 monoterpenoids, 14 phenoliacids and their glycosides, 12 steroids and organic acids.Conclusion In this study, UPLC-ESI-Q-TOF-MS/MS was used to quickly and systematically identify chemical constituents in compound Uncaria Antihypertensive Tablets. It provides the scientific theory basis for the quality control and clinical application of this drug.
