一测多评法同时测定金母颗粒中6种成分 被引量:1

Simultaneous Determination of 6 Components in Jinmu Granules with Multi-components by Single Marker

摘要 目的建立一测多评法同时测定金母颗粒(金刚刺、土茯苓、火炭母等)中没食子酸、绿原酸、虎杖苷、落新妇苷、白藜芦醇、盐酸小檗碱等6种成分的含量。方法采用高效液相法分析,Agilent ZORBAX SB-C18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.1%磷酸,梯度洗脱;体积流量1.0 mL·min^(-1);柱温35℃;检测波长291 nm;进样量10μL。以盐酸小檗碱为内标物,计算其他5种成分的相对校正因子,测定其含量。同时通过外标法和一测多评法测定11批金母颗粒中6个成分的含量,并比较两者的差异。结果6种成分在各自范围内线性关系良好(r>0.9994),平均加样回收率为88.74%~106.03%,RSD为0.41%~2.85%,以盐酸小檗碱为内标,没食子酸、绿原酸、虎杖苷、落新妇苷和白藜芦醇的相对校正因子分别为0.8666、0.7463、0.4663、0.6591、0.2683。一测多评法所得结果与外标法接近,相对平均偏差(RAD)<5.0%。结论该方法稳定,准确可靠,可为金母颗粒的质量控制和评价提供理论参考。 Objective Multi-components by single marker method was developed for the simultaneous determination of gallic acid,chlorogenic acid,polydatin,astilbin,resveratrol and berberine hydrochloride in Jinmu Granules(Smilacis Chinae Rhizoma,Smilacis Glabrae Rhizoma,Polygonum Chinese L.etc.).Methods An HPLC method was established:Agilent ZORBAX SB-C_(18)column (250 mm×4.6 mm,5μm),acetonitrile-0.1%phosphoric acid as the mobile phase by gradient elution,a volume flow of 1.0 mL·min^(-1),column temperature at 35℃,detection wavelength at 291 nm,and an injection volume of 10μL.Using berberine hydrochloride as the internal standard,we calculated the relative correction factors of the other five components to determine the content.At the same time,the contents of the six components in 11 batches of Jinmu Granules were calculated and compared by external standard method and quantitative analysis of multi-components by single marker method.Results The linear relationships of the six components were good in their respective ranges (r>0.999 4).The average recoveries were88.74%-106.03%,RSD was 0.41%-2.85%.Berberine hydrochloride was used as internal standard,the relative correction factors of gallic acid,chlorogenic acid,polydatin,astilbin and resveratrol were 0.866 6,0.746 3,0.466 3,0.659 1 and 0.268 3,respectively.The results of multi-components by single marker method showed good agreement with those of external standard method,the relative mean deviation (RAD) was less than 5.0%Conclusion The method is stable,accurate and reliable.The study can provide theoretical reference and support for quality control and evaluation of Jinmu Granules.
