子宫内膜癌患者血清和癌组织中LncRNA CRNDE、miR-153-5p表达及临床诊断价值

Expression levels of lncRNA CRNDE and miR-153-5p in serum and cancer tissue of patients with endometrial cancer and their diagnostic potential

摘要 目的 探讨miR-153-5p、LncRNA CRNDE在子宫内膜癌(EC)患者血清和癌组织中的表达及对EC的诊断价值。方法 选取2019年12月至2021年12月手术治疗的115例EC患者为观察组,收集手术患者的癌组织及癌旁组织,同时选取100例体检健康者为对照组。实时荧光定量PCR(qRT-PCR)法检测血清和癌组织中miR-153-5p、LncRNA CRNDE表达水平,分析血清中miR-153-5p、LncRNA CRNDE表达水平与患者临床病理参数的关系;受试者工作特征(ROC)曲线评价血清LncRNA CRNDE、miR-153-5p表达水平对EC发生的诊断价值。结果 与癌旁组织相比,LncRNA CRNDE在癌组织中的表达水平升高,miR-153-5p表达水平降低,且二者呈负相关(r=-0.268,P<0.05);与对照组比较,LncRNA CRNDE在观察组患者血清中的表达水平升高,miR-153-5p表达水平降低,且二者呈负相关(r=-0.315,P<0.05)。EC患者血清中LncRNA CRNDE、miR-153-5p表达水平均与年龄、肿瘤直径、组织类型无关(P>0.05),均与肌层浸润深度、分化程度、淋巴结转移、FIGO分期有关(P<0.05);ROC曲线结果显示,血清miR-153-5p、LncRNA CRNDE表达水平诊断EC发生的曲线下面积分别为0.901(95%CI:0.855~0.936)、0.944(95%CI:0.906~0.970),对应的敏感度分别为90.43%、87.83%,特异度分别为76.52%、92.17%,LncRNA CRNDE、miR-153-5p联合诊断的曲线下面积为0.971(95%CI:0.940~0.988),敏感度为89.57%,特异度为98.26%。结论 EC患者癌组织及血清中miR-153-5p呈低表达,LncRNA CRNDE呈高表达,二者联合检测对EC的诊断效能较高,可以为临床早期诊断提供参考依据。 Objective To measure the expression levels of lncRNA CRNDE and microRNA-153-5p(miR-153-5p)in serum and cancer tissues of endometrial cancer(EC)patients and their diagnostic potential.Methods A total of 115 EC patients who were surgically treated in our hospital from December 2019 to December 2021 were recruited in the observation group.Their cancer tissues and paracancerous tissues were collected.Totally 100 healthy subjects during the same period were included in the control group.The expression levels of lncRNA CRNDE and miR-153-5p in serum and cancer tissues were detected by quantitative real-time polymerase chain reaction(qRT-PCR).Their correlation with clinicopathological parameters of EC patients was analyzed.The diagnostic potential of serum lncRNA CRNDE and miR-153-5p in EC was evaluated by plotting the receiver operating characteristic(ROC)curve.Results Compared with those of paracancerous tissues,lncRNA CRNDE was upregulated and miR-153-5p was downregulated in EC tissues,showing a negative correlation(r=-0.268,P<0.05).Compared with the control group,serum level of lncRNA CRNDE was significantly higher in the observation group,while that of miR-153-5p was lower,showing a negative correlation(r=-0.315,P<0.05).Expression levels of lncRNA CRNDE and miR-153-5p in serum of EC patients were not correlated with age,tumor diameter and histological type of EC patients(P>0.05),while the above indexes were significantly correlated with the depth of myometrial invasion,degree of differentiation,lymph node metastasis and the Federation Internationale de Gynecologie et d’Obstetrique(FIGO)staging(all P<0.05).ROC curves showed that the area under the curve(AUC)of serum miR-153-5p and lncRNA CRNDE in diagnosing EC was 0.901(95%CI∶0.855,0.936)and 0.944(95%CI∶0.906,0.970),respectively.The sensitivity was 90.43%and 87.83%,and the specificity was 76.52%and 92.17%,respectively.AUC of the combination detection of lncRNA CRNDE and miR-153-5p in diagnosing EC was 0.971(95%CI∶0.940,0.988),with the sensitivity and specificity of 89.57%and 98.26%,respectively.Conclusion MiR-153-5p is downregulated,while lncRNA CRNDE is upregulated in serum and cancer tissues of EC patients.The combination detection of them presents a high diagnostic potential that provides references for the early diagnosis of EC.
