lncRNA H19、hBD-3、Survivin在OSF癌变组织中的表达及意义

摘要 目的 分析lncRNA H19、人β-防御素-3(hBD-3)、Survivin在口腔黏膜下纤维性变(OSF)癌变组织中的表达及意义。方法 选取2019年2月至2021年10月在我院接受治疗的OSF患者80例作为OSF组,另外选取OSF癌变的患者90例作为OSF癌变组,同期选取在我院进行口腔健康检查的70例健康人作为对照组,采用免疫组化、实时荧光定量法检测lncRNA H19、hBD-3、Survivin表达,分析lncRNA H19、hBD-3、Survivin在OSF癌变过程中的表达变化及三者相关性。结果 与对照组比较,OSF组、OSF癌变组中lncRNA H19、hBD-3、Survivin表达量升高(P<0.05)。与OSF组比较,OSF癌变组中lncRNA H19、hBD-3、Survivin表达量升高(P<0.05)。lncRNA H19、hBD-3、Survivin表达与OSF癌变患者肿块大小与癌变部位无关,与TNM分期、分化程度、淋巴结转移、浸润深度表达相关,Ⅲ期、Ⅳ期、中低分化、有淋巴结转移、≥1/2肌层浸润OSF癌变患者lncRNA H19、hBD-3、Survivin表达升高(P<0.05)。ROC曲线显示,与lncRNA H19、hBD-3、Survivin单项诊断相比,三项联合对OSF癌变的预测价值较高(P<0.05)。lncRNA H19、hBD-3、Survivin之间Pearson相关性分析显示,lncRNA H19与hBD-3呈正相关(r=0.513,P=0.001);lncRNA H19与Survivin呈正相关(r=0.563,P=0.001);hBD-3与Survivin呈正相关(r=575,P=0.001)。结论 lncRNA H19、hBD-3、Survivin在OSF癌变组织中表达升高,并随着疾病的演进,lncRNA H19、hBD-3、Survivin表达升高加剧,参与疾病的演进,临床上可根据其表达预测此病。 Objective To analyze the expression levels of lncRNA H19,humanβdefensin-3(hBD-3)and Survivin in oral submucosal fibrosis(OSF)cancerous tissues and their clinical significance.Methods From February 2019 to October 2021,OSF patients(OSF group,n=80),patients with OSF cancerous tissues(OSF cancerous group,n=90),healthy volunteers(control group,n=70)admitted to our hospital were enrolled.immunohistochemistry(IHC)and quantitative reverse transcription polymerase chain reaction(qRT-PCR)were performed to measure the expression levels of lncRNA H19,hBD-3 and Survivin.Their correlation during the cancerous process of OSF was assessed.Results Expression levels of lncRNA H19,hBD-3,and Survivin in the OSF group and OSF cancerous group were significantly higher than those of control group(P<0.05),which were significantly higher in the OSF cancerous group than those of OSF group(P<0.05).Expression levels of lncRNA H19,hBD-3,and Survivin were not correlated with tumor size and tumor site,while they were significantly correlated with TNM staging,differentiation degree,lymph node metastasis,and invasion depth.OSF patients with cancerous tissues who were in stageⅢ~Ⅳ,poorly differentiated,accompanied lymph node metastasis or myometrial infiltration≥1/2 had significantly upregulated lncRNA H19,hBD-3,and Survivin(P<0.05).ROC curves revealed that the predictive potential of the combined detection of lncRNA H19,hBD-3,and Survivin was significantly higher than that of the single detection(P<0.05).Pearson correlation analysis displayed that LncRNA H19 was positively correlated with hBD-3(r=0.513,P=0.001),and Survivin(r=0.563,P=0.001).hBD-3 was positively correlated with Survivin(r=575,P=0.001).Conclusion LncRNA H19,hBD-3,and Survivin are upregulated in OSF cancerous tissues,which are gradually upregulated with the disease progression.They can be served as predictive factors for OSF cancerous.
