添加低聚果糖对慢性肾脏病小鼠肠道菌群与肾脏功能的影响

摘要 目的:分析慢性肾脏病小鼠的肠道菌群的组成和丰度,探究低聚果糖对慢性肾脏病进展的影响。方法:应用经典的5/6肾切除小鼠构建慢性肾脏病模型,使用低聚果糖对慢性肾脏病小鼠进行干预治疗。运用16SrDNA高通量测序检测小鼠粪便中的微生物。结果:在三组小鼠的粪便样本中共检测出11个门,18个纲,49个目,87个属。三组间在门水平有7个门差异存在统计学意义。低聚果糖治疗后,群落的丰富度、多样性和遗传多样性显著改善。患有慢性肾脏病的小鼠补充低聚果糖后,血清白蛋白、体重显著增高,血清肌酐、尿素氮显著下降。结论:在慢性肾脏病小鼠的饮食中增加低聚果糖可使肠道微环境向健康的趋势发展,并且可有效改善CKD小鼠的营养状态和炎症。 Objective:To analyze the composition and abundance of intestinal flora in mice with chronic kidney disease(CKD),and explore the effect of Fructooligosaccharide on the progress of chronic kidney disease.Methods:Chronic kidney disease model was established in 5/6 nephrectomized mice,and fructooligosaccharide(FOS)was used to treat CKD mice.16SrDNA high-throughput sequencing was used to detect microorganisms in mouse feces.Results:A total of 11 phyla,18 classes,49 orders and 87 genera were detected in the fecal samples of three groups of mice.At the phylum level,there were significant differences in 7 phyla among the three groups.After treatment with fructooligosaccharide,the richness,diversity and genetic diversity of the community were significantly improved.After fructooligosaccharide supplementation,serum albumin and body weight increased significantly,serum creatinine and urea nitrogen decreased significantly in mice with chronic kidney disease.Conclusion:Increasing fructooligosaccharides in the diet of mice with chronic kidney disease can make the intestinal microenvironment develop to a healthy trend,and can effectively improve the nutritional status and inflammatory state of CKD mice.
