橄榄苦甙/葡萄糖酸氯己定复配制剂抗菌效果及机制

摘要 目的 探讨橄榄苦甙/葡萄糖酸氯己定复配制剂的抗菌效果及作用机制,为含植物成分消毒剂的应用提供依据。方法 采用橄榄苦甙与葡萄糖酸氯己定配制成复配制剂,采用抑菌环实验测定其对大肠杆菌、金黄色葡萄球菌和白色念珠菌的抑菌环直径,进行析因设计方差分析;采用碱性磷酸酶试剂盒和电导率仪分别检测复配制剂作用于三种菌株后碱性磷酸酶(AKP)和电导率的变化,并对AKP的变化进行单因素方差分析,对电导率的变化进行描述分析。结果 葡萄糖酸氯己定和橄榄苦甙/葡萄糖酸氯己定复配制剂均有抑菌作用,橄榄苦甙与葡萄糖酸氯己定对大肠杆菌和金黄色葡萄球菌存在交互作用(F值分别为35.47、11.61,P<0.01);复配制剂分别作用于三种菌株后,与对照组相比,AKP漏出量增多(F值分别为2 340.80、1 024.75、688.97,P<0.01)。复配制剂分别作用于三种菌后,与对照组相比,电导率均增高;但与单纯葡萄糖酸氯己定作用相比,电导率均下降。结论 橄榄苦甙/葡萄糖酸氯己定复配制剂抑菌活性强,可通过作用于细菌细胞壁发挥抗菌作用。 Objective To explore the antibacterial effect and mechanism of oleuropein/chlorhexidine gluconate compoundpreparation,so as to provide the basis for the application of plant-based disinfectants.Methods The compound preparation of oleuropein and chlorhexidine glucoate was prepared.Inhibitory ring diameter of the compound against Escherichia coli,Staphylococcus aureus and Candida albicans was measured by inhibitory ring test and factorial design analysis of variance was performed.Alkaline phosphatase(AKP)and electrical conductivity of the 3 strains were detected by alkaline phosphatase kit and electrical conductivity meter,and the changes of AKP were analyzed by one-way analysis of variance.The change of conductivity was describe and analyze.Results Both chlorhexidine gluconate and chlorhexidine/chlorhexidine gluconate compound had bacteriostatic effects(P<0.01).Chlorhexidine and chlorhexidine gluconate had interaction effects on Escherichia coli and Staphylococcus aureus(F values were 35.47 and 11.61,P0.01).Compared with the control group,the leakage of AKP increased after the compound preparation acted on the 3 strains respectively(P<0.01).Compared with the control group,the electrical conductivity of the 3 bacteria treated with the compound preparation was significantly higher,but it was decreased compared with the chlorhexidine gluconate alone.Conclusion The olivoprin/chlorhexidine gluconate compound preparation has strong antibacterial activity and can play an antibacterial role by acting on bacterial cell wall,which provides a basis for the research of a new disinfectant with high efficiency and low toxicity.
