IL-1β及NF-κB信号通路在颅内感染大鼠氧化应激反应中的作用及意义 被引量:3

Role and significance of IL-1β and NF-κB signaling pathways in oxidative stress in rats with intracranial infection

摘要 目的探究白细胞介素-1β(IL-1β)及核转录因子κB(NF-κB)信号通路在颅内感染大鼠氧化应激反应中的作用及意义。方法采用随机数字法将成年雄性SD大鼠分成模型组与对照组,模型组采用脂多糖诱导大鼠颅内感染模型,对照组采用相同体积生理盐水替代;采用酶联免疫吸附测定法检测大鼠血清和脑组织中IL-1β表达水平;试剂盒检测大鼠脑组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)水平;实时荧光定量聚合酶链式反应法和蛋白印迹法检测大鼠脑组织中IL-1β和NF-κB信号通路相关基因蛋白的表达量。结果与对照组相比,模型组大鼠血清和脑组织中IL-1β表达水平升高(P<0.05);脑组织中SOD、GSH、GSH-Px含量均降低(P<0.05);MDA含量升高(P<0.05);模型组大鼠血清及脑组织中IL-1β、NF-κB p65、p-NF-κB p65、IκB和p-IκB基因和蛋白表达水平较对照组相比,含量均增高(P<0.05)。结论颅内感染大鼠体内IL-1β及NF-κB通路相关基因蛋白高表达,提示IL-1β及NF-κB信号通路与颅内感染大鼠氧化应激反应密切相关,有望作为预测早期颅内感染的生化指标。 OBJECTIVE To explore the role and significance of Interleukin-1β(IL-1β) and transcription factor κB(NF-κB) signaling pathways in oxidative stress in rats with intracranial infection. METHODS Adult male SD rats were divided into the model group and control group by random number method.The model group was treated with lipopolysaccharide to induce rat intracranial infection model, and the control group was administered with the same volume of normal saline.ELIAS method was used to detect expression level of IL-1β in serum and brain of rats.The expression levels of superoxide dismutase(SOD), malondialdehyde(MDA), glutathione(GSH) and glutathione peroxidase(GSH-Px) in rat brain tissue were detected by the kit.Real-time quantitative polymerase chain reaction method and Western blot method were used to detect the expression of IL-1β and NF-κB signaling pathway-related genes and proteins in rat brain tissue. RESULTS Compared with the control group, the expression levels of IL-1β in the serum and brain tissue of the model group were increased(P<0.05);the contents of SOD, GSH and GSH-Px in the brain tissue were decreased(P<0.05) while the content of MDA increased(P<0.05).Compared with the control group, the expression levels of IL-1β, NF-κB p65, p-NF-κB p65, IκB and p-IκB gene and protein in the serum and brain tissue of the model group were significantly higher than those in the control group(P<0.05). CONCLUSION The expression levels of IL-1β and NF-κB pathway-related genes and proteins significantly increase in rats with intracranial infection, suggesting that IL-1β and NF-κB signaling pathways are closely related to oxidative stress in rats with intracranial infection, which can be biochemical markers of intracranial infection.
