摘要
目的探究化脓性乳腺炎患者外周血单个核细胞(PBMCs)中自噬标志物表达水平。方法选取2019年6月-2021年6月医院收治的化脓性乳腺炎患者81例为研究组,分析其感染病原菌分布情况;招募同期因非感染性疾病入院检查的哺乳期女性80例为对照组,采用密度梯度离心法分离PBMCs,实时荧光定量聚合酶链反应(PCR)法检测PBMCs中自噬标志物Beclin1、分子微管相关蛋白轻链3(LC3)及p62 mRNA水平。结果81例化脓性乳腺炎患者脓液标本阳性率为43.21%(35/81),共分离出病原菌55株,其中革兰阳性菌51株,占92.73%,革兰阴性菌4株,占7.27%;金黄色葡萄球菌是占比(74.55%)最高的病原菌。研究组患者PBMCs中Beclin1和LC3 mRNA水平高于对照组,p62 mRNA水平低于对照组(P<0.05);化脓性乳腺炎患者治疗后PBMCs中Beclin1和LC3 mRNA水平低于治疗前,p62 mRNA水平高于治疗前(P<0.05)。结论化脓性乳腺炎患者存在自噬过度活化的现象,检测自噬物标志物表达水平可为早期观察和治疗情况提供有效信息。
OBJECTIVE To explore expression levels of autophagy markers in peripheral blood mononuclear cells(PBMCs)of patients with suppurative mastitis.METHODS A total of 81 patients with suppurative mastitis admitted in the hospital between Jun 2019 and Jun 2021 were enrolled.The distribution of pathogens was analyzed.A total of 80 lactation women undergoing examinations caused by non-infectious diseases during the same period were enrolled as the control group.PBMCs were isolated by density gradient centrifugation.The mRNA expression levels of autophagy markers including Beclin1,microtubule-associated protein 1 light chain 3(LC3)and p62 in PBMCs were detected by real-time fluorescent quantitative polymerase chain reaction(PCR).RESULTS The positive rate of pus samples in 81 patients with suppurative mastitis was 43.21%(35/81).Total of 55 strains of pathogens were isolated,including 51 strains of gram-positive bacteria(92.73%)and 4 strains of gram-negative bacteria(7.27%).The proportion of Staphylococcus aureus was the highest in pathogens(74.55%).The levels of Beclin1 and LC3 mRNA in PBMCs in the study group were significantly higher than those in the control group,while level of p62 mRNA was significantly lower than that in the control group(P<0.05).After treatment,mRNA levels of Beclin1 and LC3 in PBMCs were decreased,while p62 mRNA was increased(P<0.05).CONCLUSION There is excessive activation of autophagy in patients with suppurative mastitis.Detection of the expression levels of autophagy markers can provide effective information for early observation and treatment.
