摘要
目的建立鲜枸杞子浆的质量标准。方法采用薄层色谱法对鲜枸杞子浆中的枸杞子进行定性鉴别;采用紫外-可见分光光度法测定枸杞多糖的含量,检测波长为490 nm;采用高效液相色谱法测定甜菜碱的含量,色谱柱为Merck Purospher@STAR NH2柱(250 mm×4.6 mm,5μm),流动相为乙腈-水溶液(85∶15,V/V),流速为1.0 mL/min,检测波长为195 nm,柱温为25℃,进样量为10μL。结果薄层色谱图中,斑点清晰,分离度好。D-无水葡萄糖的质量浓度在2.5060~18.7948μg/mL范围内与吸光度线性关系良好(r=0.9998,n=6);枸杞多糖的精密度、重复性、稳定性试验结果的RSD均低于2.0%;平均加样回收率为99.37%,RSD为0.54%(n=6)。甜菜碱的质量浓度在17.54~350.77μg/mL范围内与峰面积线性关系良好(r=0.9996,n=5);精密度、重复性、稳定性试验结果的RSD均低于2.0%;平均加样回收率为100.18%,RSD为0.53%(n=6)。结论该方法简便可行、重复性好,可用于鲜枸杞子浆的质量控制。
Objective To establish the quality standard of fresh Lycii Fructus syrup.Methods Lycii Fructus in fresh Lycii Fructus syrup was qualitatively identified by the thin-layer chromatography(TLC)method.The content of lycium barbarum polysaccharide was determined by the UV visible spectrophotometry with a detection wavelength of 490 nm.The content of betaine was determined by the high-performance liquid chromatography(HPLC)method,the chromatographic column was Merck Purospher@STAR NH2 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-aqueous solution(85∶15,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 195 nm,the column temperature was 25℃,and the injection volume was 10μL.Results TLC spots were clear and well separated.The linear range of D-glucopyranose was 2.5060-18.7948μg/mL(r=0.9998,n=6).The RSDs of the precision,repeatability,and stability tests of lycium barbarum polysaccharide were lower than 2.0%.The average recovery of D-glucopyranose was 99.37%with an RSD of 0.54%(n=6).The linear range of betaine was 17.54-350.77μg/mL(r=0.9996,n=5),the RSDs of precision,repeatability,and stability tests of betaine were lower than 2.0%,and the average recovery of betaine was 100.18%with an RSD of 0.53%(n=6).Conclusion This method is simple and feasible with good repeatability,which can be used for the quality control of fresh Lycii Fructus syrup.
