非编码RNA肌动蛋白纤维相关蛋白1-反义RNA对子宫内膜癌的诊断价值

Diagnostic value of long non-coding RNA actin filament-associated protein 1-antisense RNA in the endometrial carcinoma

目的探究长链非编码RNA(LncRNA)肌动蛋白纤维相关蛋白1-反义RNA(AFAP1-AS)在子宫内膜癌(EC)中的表达意义,并分析其对EC的诊断价值。方法选取郑州人民医院2017年1月至2022年1月收治的45例确诊的EC患者为研究对象,作为观察组,选择同期确诊为子宫肌瘤并接受子宫切除手术的50例患者为对照组。收集观察组癌组织和对照组正常子宫内膜组织,采用实时荧光定量PCR(qRT-PCR)法检测2组患者组织样本中LncRNAAFAP1-AS表达水平,分析其在不同临床病理特征癌组织中的表达情况,并采用受试者工作特征(ROC)曲线分析LncRNA AFAP1-AS对EC的诊断价值。结果EC患者癌组织中LncRNA AFAP1-AS相对表达量明显高于对照组,差异有统计学意义(t=37.106,P<0.001)。FIGOⅢ~Ⅳ期、病理Ⅲ级、肌层深浸润均为EC患者LncRNA AFAP1-AS高表达的危险因素(OR=0.172,P=0.044;OR=0.151,P=0.014;OR=0.214,P=0.040)。ROC曲线显示,LncRNA AFAP1-AS对EC具有较高诊断价值,诊断曲线下面积、敏感性、特异性和临界值分别为0.872、75.56%、92.00%、4.065。结论LncRNA AFAP1-AS与EC的发生发展以及临床病理特征具有密切联系,可为EC临床诊断和预后评估提供参考。

Objective To investigate the role of long non-coding RNA(LncRNA)actin filament-associated protein 1-antisense RNA(AFAP1-AS)in the endometrial carcinoma(EC),and th analyze its diagnostic value for EC.Methods A total of 45 patients with EC admitted to Zhengzhou people’s Hospital from January 2017 to January 2022 were selected as the research objects(the observation group),and 50 patients who were examined as uterine fibroids and underwent uterine extraction surgery during the same period were selected as the control group.The lesion tissues of the observation group and normal endometrial tissues of the control group were collected,and the expression levels of LncRNA AFAP1-AS in tissue samples collected from patients of the two groups were detected by quantitative real-time PCR(qRT-PCR),and the expression of different pathological features were analyzed.Receiver operating characteristic(ROC)curve was used to analyze the diagnostic value of LncRNA AFAP1-AS in EC.Results The relative expression of LncRNA AFAP1-AS in EC patients was significantly higher than that in the control group,the difference was statistically significant(t=37.106,P<0.001).FIGO stageⅢ-Ⅳ,pathological gradeⅢ,and muscle deep infiltration were the risk factors for high expression of LncRNA AFAP1-AS in EC patients(OR=0.172,P=0.044;OR=0.151,P=0.014;OR=0.214,P=0.040).In the ROC curve,LncRNA AFAP1-AS had high diagnostic value for EC,and the diagnostic area under curve,sensitivity,specificity and cut-off value were 0.872,75.56%,92.00%and 4.065.Conclusion LncRNA AFAP1-AS is closely related to the occurrence,development and clinicopathological features of EC,which can provide reference for the clinical diagnosis and prognosis evaluation of EC.
