桃红四物汤对急性脑缺血再灌注损伤小鼠神经功能缺损评分及脑梗死体积的影响研究

Study on the effect of Taohong Siwu Decoction on the neurological deficit score and cerebral infarction volume in mice with acute cerebral ischemia/reperfusion injury

摘要 目的分析桃红四物汤对急性脑缺血再灌注损伤(CIR)小鼠神经功能缺损评分及脑梗死体积的影响。方法90只健康小鼠,随机分为假手术组、CIR模型组、桃红四物汤组,每组30只。CIR模型组、桃红四物汤组小鼠建立急性CIR模型,假手术组只进行血管分离,不进行其他操作;桃红四物汤组小鼠给予桃红四物汤灌胃,假手术组、CIR模型组给予等体积蒸馏水灌胃。比较三组小鼠脑血流值、神经功能缺损评分、脑梗死体积及脑含水量。结果CIR 24 h、CIR 1周,CIR模型组、桃红四物汤组小鼠脑血流值均低于假手术组,但桃红四物汤组小鼠高于CIR模型组,差异具有统计学意义(P<0.05)。CIR 24 h、CIR 1周,假手术组小鼠神经功能缺损评分为0分,未见神经功能缺损;CIR 24 h、CIR 1周,桃红四物汤组小鼠神经功能缺损评分分别为(2.65±0.11)、(0.38±0.06)分,均低于CIR模型组的(3.12±0.67)分、(0.98±0.23)分,差异具有统计学意义(P<0.05)。CIR 24 h、CIR 1周,假手术组小鼠脑梗死体积均为0;CIR 24 h、CIR 1周,桃红四物汤组小鼠脑梗死面积分别为(20.63±1.59)%、(12.78±1.03)%,均低于CIR模型组的(26.75±2.14)%、(16.73±1.56)%,差异具有统计学意义(P<0.05)。CIR 24 h、CIR 1周,桃红四物汤组的脑含水量分别为(78.14±0.23)%、(77.85±0.11)%,假手术组分别为(80.59±1.02)%、(78.64±0.57)%,CIR模型组分别为(76.15±0.63)%、(76.11±0.24)%,桃红四物汤组、CIR模型组小鼠的脑含水量高于假手术组,但桃红四物汤组小鼠低于CIR模型组,差异具有统计学意义(P<0.05)。结论桃红四物汤用于小鼠急性CIR中效果显著,有利于增加脑血流值,减轻神经功能缺损程度,缩小脑梗死体积,减少脑含水量,值得临床推广应用。 Objective To analyze the effect of Taohong Siwu Decoction on the neurological deficit score and cerebral infarction volume in mice with acute cerebral ischemia/reperfusion injury(CIR).Methods A total of 90 healthy mice were randomly divided into sham operation group,CIR model group and Taohong Siwu Decoction group,with 30 mice in each group.Acute CIR model was established in CIR model group and Taohong Siwu Decoction group,and sham operation group only performed vascular separation without other operations.Taohong Siwu Decoction group was given Taohong Siwu Decoction by gavage,sham operation group and CIR model group were given equal volume of distilled water by gavage.The cerebral blood flow,neurological deficit score,cerebral infarct volume and cerebral water content were compared among the three groups.Results At 24 h and 1 week of CIR,the cerebral blood flow in CIR model group and Taohong Siwu Decoction group were lower than than those of the sham operation group,but the Taohong Siwu Decoction group was higher than the CIR group.The differences were statistically significant(P<0.05)At 24 h and 1 week of CIR,the neurological deficit scores of mice in the sham operation group were 0,and no neurological deficit was seen;at 24 h and 1 week of CIR,the neurological deficit scores of mice in the Taohong Siwu Decoction group were(2.65±0.11)and(0.38±0.06)points,which were lower than those of(3.12±0.67)and(0.98±0.23)points in the CIR model group.The differences were statistically significant(P<0.05).At 24 h and 1 week of CIR,the cerebral infarct volume of mice in the sham operation group was 0.At 24 h and 1 week of CIR,the cerebral infarct area of mice in the Taohong Siwu Decoction group were(20.63±1.59)%and(12.78±1.03)%,which were lower than those of(26.75±2.14)%and(16.73±1.56)%in the CIR model group,and the differences were statistically significant(P<0.05).At 24 h and 1 week of CIR,the cerebral water content were(78.14±0.23)%and(77.85±0.11)%in Taohong Siwu Decoction group,(80.59±1.02)%and(78.64±0.57)%in the sham operation group,and(76.15±0.63)%and(76.11±0.24)%in the CIR model group.The cerebral water content of mice in the Taohong Siwu Decoction group and CIR model group was higher than that in the sham operation group,but the Taohong Siwu Decoction group were lower than the CIR model group.The differences were statistically significant(P<0.05).Conclusion Taohong Siwu Decoction has a significant effect on the neuroprotection of acute CIR in mice,which is beneficial to increase cerebral blood flow,reduce the degree of neurological defects,reduce the cerebral infarct volume,and reduce the cerebral water content.It is worthy of clinical promotion and application.
