摘要 目的探讨尖尾芋石油醚部位的化学成分及其抗乳腺癌作用。方法采用GC-MS法分析尖尾芋石油醚部位(EAC)的化学成分,CCK-8法检测EAC对MDA-MB-231细胞和4T1细胞活性的影响,显微镜观察EAC对MDA-MB-231形态的影响,Western blot法检测EAC处理后,MAPKs信号通路蛋白的表达情况。结果共鉴定出53个化学成分,主要成分为脂肪酸类、甾醇类等。EAC能抑制MDA-MB-231细胞和4T1细胞的活性,改变MDA-MB-231细胞的形态,能抑制MAPKs信号通路中p-ERK/ERK的表达。结论EAC可抑制MDA-MB-231乳腺癌细胞的增殖,可能与下调ERK蛋白磷酸化有关。 Objective To determine the chemical compositions and mechanism of anti-breast cancer activity of petroleum ether site of Alocasia cucullate(Lour.)Schoot.(EAC).Methods The chemical compositions in EAC were analyzed by GC-MS,the effect of EAC on the activity of MDA-MB-231 cells and 4T1 cells was detected by CCK-8,the morphology of MDA-MB-231 cells was observed by microscopy,and the level of protein in MAPKs signaling pathway was detected by Western blot.Results Totally 53 chemical components being identified,the major components were fatty acids and sterols.The activity of MDA-MB-231 cells and 4T1 cells and the expression of p-ERK/ERK protein in the MAPKs signaling pathway were inhibited by EAC.The morphology of MDA-MB-231 cells was changed by EAC.Conclusion EAC can inhibit the proliferation of MDA-MB-231 cells,possibly by down-regulating the phosphorylation of ERK protein.
