二陈汤治疗多囊卵巢综合征的网络药理学机制

The network pharmacological mechanism of the Erchen decoction in the treatment of polycystic ovary syndrome

目的:通过网络药理学方法探究二陈汤治疗多囊卵巢综合征(Polycystic Ovary Syndrome,PCOS)的作用机制。方法:利用中药系统药理学数据库与分析平台(TCMSP)获取二陈汤的有效成分和作用靶点,通过人类基因数据库(Gene Cards)筛选出PCOS的靶点,运用Cytoscape软件构建“药物-活性成分-靶点-疾病”网络,通过STRING平台构建靶标蛋白质-蛋白质相互作用网络,并利用R包Cluster Profiler进行基因本体论(GO)、疾病本体论(DO)及京都基因和基因百科全书(KEGG)富集分析。结果:获得二陈汤与PCOS的药物-疾病靶点164个,潜在的活性成分包括槲皮素、山柰酚、柚皮素等。关键靶点有JUN激酶、信号传导及转录激活蛋白3、丝裂原活化蛋白激酶3等。富集分析显示白细胞介素-17信号通路、肿瘤坏死因子信号通路、缺氧诱导因子-1信号通路可能是二陈汤治疗PCOS的关键通路。结论:二陈汤有效成分可能通过抗炎性反应、改善胰岛素抵抗、促进卵泡发育、调节脂质代谢、调节激素水平等药理作用来治疗PCOS。

Objective:To explore the mechanism of the Erchen decoction(二陈汤)in the treatment of polycystic ovary syndrome(PCOS)through network pharmacology method.Methods:Using TCMSP database,we collected the active ingredients and targets of the Erchen decoction.The targets of PCOS were screened out through GeneCards databases.The“drug-component-target-disease”network was constructed using Cytoscape software.The target protein-protein interaction network was constructed through the STRING website,and the GO,DO as well as KEGG analysis on common targets were performed using R package clusterProfiler.Results:A total of 164 drug-disease common targets of the Erchen decoction in the treatment of PCOS were screened.Potential active ingredients include quercetin,kaempferol and naringenin.The core targets include JUN,STAT3 and MAPK3.The enrichment analysis showed that IL-17 signaling pathway,TNF signaling pathway and HIF-1 signaling pathway might be the key pathways for the treatment of PCOS with the Erchen decoction.Conclusion:The active ingredients of the Erchen decoction may treat PCOS through anti-inflammatory response,improvement of insulin resistance,promotion of follicular development and regulation of lipid metabolism as well as hormone levels.
