石氏温经宣痹通络汤对小鼠膝骨关节炎软骨组织的保护作用

The protective effect of the SHI’s Wenjing Xuanbi Tongluo decoction on cartilage tissue of knee osteoarthritis mice

目的:观察石氏温经宣痹通络汤对膝骨关节炎小鼠软骨组织的影响。方法:将40只无特定病原体级C57BL雄性小鼠随机分为正常组、模型组、中药组、西乐葆组,每组10只,采用内侧半月板失稳法建立膝骨关节炎模型。每组分别给予相应的药物。治疗周期结束后,取小鼠膝关节软骨进行苏木素-伊红染色剂和番红固绿染色剂染色,光镜下观察软骨组织学变化和类胰岛素生长因子1(Insulin-like Growth Factor 1,IGF-1)分布情况,进行染色强度的积分吸光度图像分析。结果:光镜下显示,中药组小鼠软骨结构基本完整,透明软骨无明显纤维化,关节软骨病理积分低于模型组,差异有统计学意义(P<0.05),而中药组与西乐葆组的差异无统计学意义(P>0.05);免疫组化分析:中药组IGF-1染色强度的积分吸光度高于模型组,差异有统计学意义(P<0.05);中药组IGF-1染色强度的积分吸光度与西乐葆组比较,差异无统计学意义(P>0.05)。结论:石氏温经宣痹通络汤具有修复软骨损伤、阻止透明软骨纤维化、保持与恢复软骨细胞结构完整性的作用,可用于治疗小鼠膝骨性关节炎,但作用并不优于塞来昔布胶囊。

Objective:To observe the influence of the SHI’s Wenjing Xuanbi Tongluo decoction(石氏温经宣痹通络汤)on cartilage tissue of knee osteoarthritis mice.Methods:Forty specific pathogen free C57BL male mice were randomly divided into the normal group,the model group,the TCM group and the celebrex group,with 10 mice in each group.The knee osteoarthritis model was established by destabilization of medial meniscus method.Each group was given corresponding drugs respectively.At the end of the treatment cycle,the knee cartilage tissue of knee osteoarthritis mice was stained by hematoxylin-eosin staining and safranin fast green staining.The histological changes of cartilage,and the distribution of IGF-1 were observed under light microscope,and the integral absorbance image analysis of staining intensity was performed finally.Results:Under the light microscope,the cartilage structure in the TCM group was basically intact,the hyaline cartilage had no obvious fibrosis,and the pathological score of articular cartilage in the TCM group was significantly lower than the model group,and the difference was statistically significant(P<0.05).There was no significant difference between the TCM group and the celebrex group(P>0.05).The immunohistochemical analysis showed that the integral absorbance of IGF-1 staining intensity in the TCM group was higher than the model group,and the difference was statistically significant(P<0.05).There was no significant difference in the integral absorbance of IGF-1 staining intensity between the TCM group and the celebrex group(P>0.05).Conclusion:The SHI’s Wenjing Xuanbi Tongluo decoction has the functions of repairing cartilage damage,preventing hyaline cartilage fibrosis,maintaining and restoring the structural integrity of cartilage cells,and can be used to treat knee osteoarthritis in mice,but its effect is not better than celecoxib capsules.
