基于HPLC-QAMS多指标成分联合化学计量学及熵权TOPSIS的白及综合质量评价 被引量:3

Comprehensive Quality Evaluation of Bletillae Rhizoma Based on Multi-index Components by HPLC-QAMS Combined with Chemometrics and EW-TOPSIS

摘要 目的采用高效液相色谱-一测多评法(HPLC-QAMS)联合化学计量学及熵权TOPSIS(EW-TOPSIS)对白及质量进行评价。方法收集全国5省18批白及样品,以1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯为参照物,同时测定白及中天麻素、4-(葡糖糖氧基)-肉桂酸葡萄糖氧基苄酯、1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯-2-葡萄糖苷、手参苷Ⅲ、手参苷Ⅸ、1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯、贝母兰宁、2,7-二羟基-3,4-二甲氧基菲、白及联菲A、白及联菲B、山药素Ⅲ和3'-O-甲基山药素Ⅲ的含量,运用化学计量学方法对含量测定结果进行综合分析,挖掘影响白及产品质量的主要潜在标志物,并对不同产地来源白及进行质量优劣性评价。结果12种成分分别在各自范围内线性关系良好,准确度良好;HPLC-QAMS和外标法含量测定结果差异无统计学意义(P>0.05);化学计量学方法显示18批白及样品可聚为3类,呈现一定的产区差异;山药素Ⅲ、1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯、天麻素和贝母兰宁是影响白及产品质量的主要潜在标志物;EW-TOPSIS分析结果显示5个省中贵州和四川地区白及样品质量最优,其次为湖北、江西和陕西。结论本研究建立的HPLC-QAMS方法操作便捷、结果准确,化学计量学及EW-TOPSIS方法可用于白及质量的综合评价。 Objective To establish a method for the quality evaluation of Bletillae Rhizoma by HPLC-QAMS combined with chemometrics and entropy weight TOPSIS method.Methods A total of 18 samples from 5 provinces were collected,and militarine was used as the reference substance.The contents of gastrodin,shancigusin I,dactylorhin A,gymnosideⅢ,gymnosideⅨ,militarine,coelonin,2,7-dihydroxy-3,4-dimethoxyphenanthrene,blestriarene A,blestriarene B,batatasinⅢand 3'-O-MethylbatatasinⅢwere determined simultaneously in Bletillae Rhizoma.Chemometrics method were used to comprehensively analyze the content determination results,and the main potential markers affecting the quality of Bletillae Rhizoma were excavated.The quality of Bletillae Rhizoma from different origins was evaluated.Results The 12 components had a good linear relationship in their respective ranges,and accuracy was good.There was no significant difference in the determination results of HPLC-QAMS method and external standard method(P>0.05).The chemometrics method showed that 18 batches of Bletillae Rhizoma could be clustered into three categories,showing certain regional differences.BatatasinⅢ,militarine,gastrodin and coelonin were the main potential markers affecting the quality of Bletillae Rhizoma.The entropy weight TOPSIS method demonstrated that the quality of Bletillae Rhizoma in Guizhou Province and Sichuan Province was the best in all provinces,followed by Hubei,Jiangxi and Shaanxi provinces.Conclusion The established HPLC-QAMS method is convenient and accurate,and chemometrics combined with EW-TOPSIS method can be used for the comprehensive evaluation of the quality of Bletillae Rhizoma.
