中药复方柴枳导滞汤对糖尿病模型大鼠糖脂代谢与瘦素的影响

Effects of Chaizhi Daozhi Decoction on Glucose and Lipid Metabolism Indexes and Leptin in Diabetic Rats

摘要 目的:观察中药复方柴枳导滞汤对糖尿病模型大鼠体质量、糖脂代谢、瘦素等指标和舌乳头病理形态的影响。方法:SPF级雄性SD大鼠40只,随机取8只作为空白组,给予普通饲料喂养,余下采用“高糖高脂喂养4周+链脲佐菌素(STZ)注射”方法建立糖尿病大鼠模型,造模成功的大鼠随机分为模型组、二甲双胍组及柴枳导滞汤组并分别给药8周。记录各组大鼠体质量及尾静脉血糖,检测空腹血糖(FBG)、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、尿酸(UA)等生化指标,采用ELISA法检测胰岛素(FINS)、瘦素(LEP)、脂联素(ADP)、超氧化物歧化酶(SOD)、丙二醛(MDA)、白介素6(IL-6)及肿瘤坏死因子α(TNF-α),计算ISI和HOMA-IR,HE染色法观察舌乳头病理形态。结果:与空白组相比,模型组大鼠体质量下降,FBG、TG、TC、UA、FINS、LEP、MDA、IL-6、TNF-α水平及HOMA-IR升高(P<0.05),HDL-C、ADP、SOD水平及ISI降低(P<0.05),LDL-C水平无明显变化(P>0.05);与模型组相比,柴枳导滞汤组大鼠体质量下降缓慢,FBG、TC、UA、FINS、LEP、TNF-α水平及HOMA-IR降低(P<0.05),HDL-C、ADP、SOD水平及ISI升高(P<0.05),与二甲双胍组相比指标改善程度差异无统计学意义(P>0.05)。结论:柴枳导滞汤能够改善糖尿病模型大鼠的糖脂代谢、减轻胰岛素抵抗、调节瘦素与脂联素水平、具有抗氧化与抗炎作用,这可能是其治疗糖尿病的作用机制之一。 Objective:To observe the effects of Chaizhi Daozhi Decoction on body weight,glucose,lipid,leptin and tongue papilla in rats with diabetic mellitus(DM).Methods:8 rats from 40 male SD rats were randomly selected into the blank group and fed with normal diet,and the remaining rats were established DM model with the method of‘high-sugar and high-fat feeding for 4 weeks plus streptozocin(STZ)injection’.The successfully modeled rats were randomly divided into the model group,the Metformin group and the Chaizhi Daozhi Decoction group,and were intervened for 8 weeks.Body weight and blood glucose from tail vein were recorded in each group.Fasting blood glucose(FBG),triglycerides(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),high-density liptein cholesterol(HDL-C),uric acid(UA),fasting serum insulin(FINS),leptin(LEP),adiponectin(ADP),superoxide dismutase(SOD),malondialdehyde(MDA),interleukin 6(IL-6)and tumour necrosis factor-α(TNF-α)were detected,and ISI and HOMA-IR were calculated.Pathological morphology of tongue papilla was observed with HE staining method.Results:Compared with those in the blank group,the body weight was decreased,the levels of FBG,TG,TC,UA,FINS,LEP,MDA,IL-6,TNF-αand HOMA-IR were increased(P<0.05),the levels of HDL-C,ADP,SOD and ISI were decreased(P<0.05),and the level of LDL-C showed no significant change in the model group(P>0.05).Compared with those in the model group,the weight loss was slowed down,the levels of FBG,TC,UA,FINS,LEP,TNF-αand HOMA-IR were decreased(P<0.05),the levels of HDL-C,ADP,SOD and ISI were increased in the Chaizhi Daozhi Decoction group(P<0.05).There were no statistical differences in the indexes improvements between the Chizhi Daozhi Decoction group and the Metformin group(P>0.05).Conclusion:Chaizhi Daozhi Decoction can improve glucose and lipid metabolism disorders,reduce insulin resistance,regulate leptin and adiponectin,and it possesses anti-oxidant and anti-inflammatory effects,which maybe one of its action mechanisms in treatment of DM.
